Polygonum cuspidatum Sieb. et Zucc. is one of traditional Chinese medicines in common use with its radix and rhizome. It is a member of Polygonum Linn.. Abundant in natural resources, this species has wide distribution in China. In this dissertation, studies on chemical constituents of the radix and rhizome of Polygonum cuspidatum are presented firstly. 17 compounds were isolated from the extraction of radix and rhizome. Among them, 14 compounds' structures were identified on the ground of physical, chemical properties and spectral data. The 14 compounds respectively are chrysophanol(1), physcion(2), emodin(3), rhein(4), reveratrol(5), piceid(6), apigenin(7), kaempferol(8), quercetin(9), β-sitosterol(10), stigmast-5-ene-3β ,7α -diol(11), daucosterol(12) , gallic acid(13) and D-mannitol(14). stigmast-5-ene-3β ,7 -diol (11) is isolated from the genus for the first time.Recently, resveratrol and piceid, aboundant in Polygonum cuspidatum, are found that they have specific pharmaceutical activities such as natable cancer chemopreventive activity, reducing the risk of coronary heart disease (CHD) and ect. Therefore, according to the new research tendency of Polygonum cuspidatum's pharmacologinical effects, we determined resveratrol and piceid as the constituents for quality evaluation of Polygonum cuspidatum, which can nearly represent the effect of Polygonum cuspidatum, .The method of simultaneous determination of resveratrol and piceid in Polygonum cuspidatum was developed for the analysis of the crude drug's quanlity by using HPLC. It is the frist time in China to establish such a method. Besides, the stability ofresveratrol and piceid was examined carefully. Moreover, the resveratrol and piceid of 25 samples were determined by the above method. The samples included the radix and rhizome of Polygonum cuspidatum of different habitats, different collecting times and different parts of this plant. It also is the first time to study the inherence quanlity of Polygonum cuspidatum relatively completely up to now. These afford reliable experimental basis for the chemistry and quanlity research on Polygonum cuspidatum in the future.
