| Objective:By digging and analyzing the key targets of Bushen Hemai Prescription,and making clinical evaluation,this research want to further explore its curative effect and possible advantages on hypertension of kidney Qi deficiency type,to provide ideas for further study on the mechanism of Bushen Hemai Prescription in preventing and treating hypertension,and to optimize the effect of TCM in lowering blood pressure.Method:Based on the chemical constituents-target network and target-disease network of Bushen Hemai Prescriptions,the key targets were excavated and the data were analyzed statistically.On the basis of previous experimental verification,clinical randomized controlled trials were used to evaluate the efficacy.A double-blind controlled study was conducted in 213 hypertensive subjects aged 50-75 years old,who were selected for the syndrome of kidney-qi deficiency.The control group was treated with placebo plus standardized antihypertensive treatment,while the experimental group was treated with BushenHemai Prescription combined with standardized antihypertensive treatment.The treatment period was 6 months.The related indicators of the two groups were observed and compared.Finally,SPSS 19.0 statistical software was used to analyze the data,and draw conclusions.Result:In the field of network target research,171 drug component targets and 824 hypertension targets were found.57 genes were obtained from the intersection.They were input into DAVID database for GO function enrichment and KEGG pathway enrichment.Finally,25 GO and 5 KEGG results were obtained.In terms of clinical efficacy evaluation,both the control group and the experimental group had antihypertensive effect,and the experimental group had more obvious effect.The experimental group and the control group measured systolic blood pressure(SeSBP),pulse pressure(SePP),and 24-hour ambulatory blood pressure monitoring measured 24hSBP,dSBP,nSBP,24hPP were significantly different from those before treatment.SeSBP,SePP,24hSBP,dSBP and 24PP decreased significantly among the groups,but there was no significant difference among the other blood pressure indicators.After treatment,the main symptoms,secondary symptoms and total scores of the two groups were decreased compared with those before treatment,and there were statistical differences between the main symptoms and total scores after treatment.The data of neck color Doppler ultrasound and heart color Doppler ultrasound analysis showed that both groups improved after treatment,but there was no significant difference between the two groups.Although the urinary microalbumin of the two groups decreased after treatment,there was no significant difference between the two groups.There was significant difference in the endogenous creatinine clearance rate between the two groups after treatment.Conclusion:According to the results of the study,we can infer that the active ingredients in Bushen Hemai Prescription can act on multiple signaling pathways by interfering with immune inflammation,regulating vasomotor and endothelial function,and so on,so as to achieve the goal of lowering blood pressure.Compared with placebo plus standardized antihypertensive drugs,Bushen Hemai Prescription combined with standardized antihypertensive drugs can significantly reduce systolic blood pressure and pulse pressure in patients with isolated systolic hypertension of grade Ⅰ and Ⅱ,and will not significantly reduce diastolic blood pressure.It can effectively improve the blood pressure compliance rate of patients with hypertension of deficiency of kidney-qi,reduce the damage of heart and kidney,improve the symptoms of patients and enhance the quality of life.To sum up,Bushen Hemai Prescription is an effective prescription for treating hypertension of kidney Qi deficiency type.It can be widely used in clinic. |
