Study On Effective Components And Mechanism Of Dachaihu Decoction In Treatment Of Acute Pancreatitis(syndrome Of Liver Depression And Qi Stagnation)

Objective To study the efficacy mechanism and material basis of Dachaihu Decoction(DCHD)in the treatment of acute pancreatitis(AP)with liver depression and qi stagnation,to provide an experimental basis for the secondary development and rational application of DCHD.Methods First,the network pharmacology was used to predict and analyze DCHD’s treatment of liver depression and qi stagnation type AP,and meta-analysis was used to conduct literature research to initially predict the mechanism and material basis of the effect of DCHD.Then build a combined model of liver depression and qi stagnation AP rats with disease and syndrome,under the combined model of disease and syndrome,in-depth study of the efficacy of DCHD.Finally,the primary cells of the pancreatic acinar of AP rats with liver depression and qi stagnation were isolated and cultured.The primary cells were used to predict the efficacy of the active ingredients.Results The results of literature studies show that the application of DCHD in the treatment of liver depression and stagnation type AP can significantly improve the clinical efficacy,and no serious adverse reactions and complications occur,which provides a clinical application basis for the subsequent study of the efficacy and basis of DCHD.Internet pharmacology research found that 83 active ingredients such as quercetin,luteolin,kaempferol,baicalein,etc.in DCHD act on 18 targets such as IL6,VEGFA,TNF,EGF,PTGS2,etc.Efficacy,the meta-analysis found that the chemical components of quercetin,baicalin,emodin,paeoniflorin,β-carotene 5 in DCHD have a significant effect on the rat model of acute pancreatitis;comprehensive consideration After the Degree value and SCURA value of each chemical component,we finally selected quercetin,luteolin,kaempferol,baicalein,baicalin,emodin,paeoniflorin,baicalin,saikosaponin a,saikosaponin d 10 active ingredients were studied in depth.In vivo,experimental results show that D DCHD can significantly reduce the syndrome of liver depression and qi stagnation type AP rats combined with the rat model syndrome score,improve rat signs,reduce serum amylase(AMY)activity and pancreatic tissue TNF-α,IL-The expression of 6 other inflammatory factors.The results of in vitro experiments found that 7 components of quercetin,baicalin,baicalin,luteolin,paeoniflorin,emodin,and saikosaponin a can significantly reduce the primary cell supernatant of pancreatic acinar in AP rats AMY activity and TNF-α and IL-6m RNA levels in cells.Conclusion DCHD exerts the efficacy of treating liver depression and qi stagnation type AP by reducing the syndrome scores of liver depression and stagnation syndrome in rats and improving the pathological damage of the pancreas.Reduces inflammation of the pancreas.The seven active ingredients of quercetin,baicalein,baicalin,luteolin,paeoniflorin,emodin,and saikosaponin a in DCHD may be the main functional substance basis.
