| ObjectiveBy detecting the changes of IL-17 concentration in gingival crevicular fluid of different types of periodontitis and periodontal healthy people,analyzing the relationship between IL-17 and various clinical indexes of different types of periodontitis,evaluating whether IL-17 in gingival crevicular fluid can be an objective index reflecting different types of periodontitis related diseases,and exploring the role of IL-17 in gingival crevicular fluid.Methods1.ObjectivesFrom January 4,2018 to October 29,2018,63 patients were selected from the Department of Stomatology of Chifeng Tumor Hospital,including 21 periodontal healthy people,who were divided into 11 men and 10 women,with the age range of 30-42 years old and the average age of 38.36 years old,and they were set up as group A;42 patients were diagnosed with periodontitis,including 21 patients who were determined as moderate speed progressive periodontitis according to the new periodontitis classification standard,who were divided into 9 men and 12 women,the age range is 38-44 years old,the average age is 40.25 years old,which is set up as group B;there are 21 patients identified as rapid progressive periodontitis,divided into 12 men and 9 women,the age range is 25-33 years old,the average age is 29.02 years old,which is set up as group C.All subjects were selected six index teeth(11,16,26,31,36,46).The selected patients were informed of the purpose and process of the trial and voluntarily participated in the trial.2.Data acquisitionTo examine the periodontal condition of every index tooth(except the third molar)and record the result in detail.Including the periodontal indexes,the probing depth(PD),the attachment loss(AL),the plaque index(PLI),and the panoramic radiography.3.Gingival crevicular fluid collectionThe tip of No.30 hygroscopic paper was cutted 4.5mm of the tip by sterile scissors and sterilized.Every four paper points for a group putted into the EP tube has been completed disinfection,and then recorded the number and weigh with an electronic balance.Cleaning the tooth surface and blow dry and scraping off the plaque on the gingival lto collect the gingival crevicular fluid wear a sterile gloves.The paper tip of the damp paperwastaked out and inserted into the gum pocket at the selected points(near Cheek,far cheek,near tongue and far tongue),stopped inserting when feel resistance.Repeat the procedure and remove the gingival crevicular fluid after 14 minutes.To record the number and the weight of the 4 hygroscopic paper tips in the EP tube,and then sealed the EP tube in-70 °C refrigerator.The sample to be tested by defrosting,adding buffer,shocking,centrifugationat room temperature,and then to be taken of the supernatant for testing.4.IL-17 level tested by Enzyme-linked immunosorbent assay.Results1.The concentrations of IL-17 in gingival crevicular fluid of the normal control group,chronic periodontitis group and invasive periodontitis group were 326.42±34.03pg/μl,425.05±63.02pg/μl,459.03±53.11pg/μl respectively.2.The concentration of IL-17 in gingival crevicular fluid of patients with chronic periodontitis was higher than that of Normal Control Group(P<0.01),and was positively correlated with PD(r=0.903,P<0.01),the positively correlated with Al(r=0.931,P<0.01),but was positively correlated with PLI(r=0.843,P<0.01).3.The concentration of IL-17 in gingival crevicular fluid of patients with invasive periodontitis was higher than that of Normal Control Group(P<0.01)and there was a positive correlation between IL-17 and PD(r=0.834,P<0.01),the positively correlated with PLI(r=0.793,P<0.01).There was no significant difference between invasive periodontitis group and chronic periodontitis group(P>0.05).Conclusions1.IL-17 can be detected in gingival crevicular fluid of healthy periodontal subjects,moderate and rapid periodontitis subjects.2.The concentration of IL-17 in gingival crevicular fluid of patients with periodontitis was higher than that control group,and the concentration of IL-17 increased with the depth of periodontal probing and the degree of attachment loss.It is speculated that the concentration of IL-17 is related to the occurrence and development of periodontitis.3.There was no significant difference in the concentration of IL-17 in gingival crevicular fluid between the two types of periodontitis. |
