Study On The Expression Of MiRNA-2861 In Follicular Fluid Of Patients With The Phlegm Dampness Polycystic Ovary Syndrome

Objective: To research the reproductive endocrine,glycolipid metabolism,controlled ovarian hyperstimulation outcome and the differential expression of miRNA-2861 in the follicular fluid in patients with the phlegm dampness PCOS,and to explore the pathogenesis of the phlegm dampness PCOS and the effect of phlegm-dampness syndrome on oocyte and embryo development.Methods: Three groups including the phlegm dampness PCOS group,the non-phlegm dampness PCOS group and the normal control group with normal ovarian reserve function were enrolled to detect the differential expression of miR-2861 with RT-q PCR in the follicle fluid.Results:(1)BMI and WHR in the phlegm dampness PCOS group were higher than that in the non-phlegm dampness PCOS group and the normal control group(P<0.01).(2)The level of T,TC,TG,LDL-C,HOMA-IR in the phlegm dampness PCOS group was higher than that in the non-phlegm dampness PCOS group and the normal control group(P<0.01),HDL-C level in the phlegm dampness PCOS group was lower than that in the non-phlegm dampness PCOS group and the normal control group(P<0.01),LH level in the phlegm dampness and the non-phlegm dampness PCOS groups was increased than that in the normal control group(P<0.01),LH level in the non-phlegm dampness PCOS group was increased than that in the phlegm dampness PCOS group(P < 0.01).(3)The oocyte retrieval rate,fertilization rate and high-qualified embryo rate of the phlegm dampness PCOS group were lower than that of the non-phlegm dampness PCOS group and the normal control group(P<0.01).(4)The oocyte retrieval rate was negatively correlated with the level of BMI,WHR,T,TC,TG,LDL and HOMA-IR(P<0.05),positively correlated with the level of HDL and LH(P<0.05);the fertilization rate was negatively correlated with the level of BMI,TC,LDL and HOMA-IR(P<0.05);the high-qualified embryo rate was negatively correlated with the level of BMI,WHR,T,TG,LDL and HOMA-IR(P<0.05),positively correlated with the level of HDL(P<0.05).(5)The expression of miR-2861 in the phlegm and the non-phlegm dampness PCOS groups was higher than that in the normal control group(P<0.05),but the expression of miR-2861 in the phlegm dampness PCOS group was lower than that in the non-phlegm dampness PCOS group(P<0.05).(6)Mi R-2861 expression in follicular fluid of PCOS patients was negatively correlated with the level of BMI,WHR,TC,TG,LDL and HOMA-IR(P<0.05),positively correlated with the level of HDL,the oocyte retrieval rate,the fertilization rate and the high-qualified embryo rate(P<0.05).Conclusion: Patients with the phlegm dampness polycystic ovary syndrome are more prone to abdominal obesity,glucose and lipid metabolism and endocrine disorders,while the outcome of controlled ovarian hyperstimulation is general.The outcome of controlled ovarian hyperstimulation was connected with the level of BMI,WHR,T and glucose and lipid metabolism disorder.Mi R-2861 was higher expressed in the follicular fluid of PCOS patients,miR-2861 was lower expressed in the follicular fluid of the phlegm dampness syndrome patients,miR-2861 may be involved in the pathogenesis of the phlegm dampness PCOS patients and the process of oocyte and embryo development.
