Experiment 1:The experimental study of the effect of PRP on the survival of adipose tissue transplantationObjective:To investigate the absorption and survival rate of PRP combined with granular autologous fat transplantation.Methods:Healthy Nude mouse were selected as the research objects.PRP and inguinal granular lipid cup were prepared and injected into the back subcutaneous of Nude mouse.Six months after transplantation,the mouse were executed.The expression of vascular endothelial growth factor was detected by HE,immunohistochemical staining and Western Blot.Protein expression.Results:(1)Scanning electron microscopy analysis showed that the structure of fat granules was intact after transplantation.(2)Observation results:At 1 month and 6 months after transplantation,the survival rate of granular fat in PRP/adipose group was higher than that in saline/adipose group.(3)HE staining:There were large vacuoles wrapped by fibrous tissue and intact neovascularization around transplanted fat in both groups.The number of neovascularization in PRP/adipose group was significantly higher than that in saline/adipose group.(4)Immunohistochemical staining:6 months after transplantation,the number of neovascularization increased in both saline/fat group and PRP/fat group,and the number of neovascularization in PRP/fat group was significantly more than that in saline/fat group.Conclusions:PRP can obviously promote the survival rate and vascularization of adipose tissue transplantation.Experiment 2:Clinical study of the effect of PRP combined with granular autologous fat transplantation on facial rejuvenation in early stageObjective:To investigate the effect of PRP combined with granular autologous fat transplantation on facial rejuvenation in early stage.Methods:58 patients who underwent autologous fat granule transplantation in our hospital were randomly divided into control group and observation group.The control group was treated with autologous fat granule transplantation only,while the observation group was treated with PRP+fat granule.Results:(1)The therapeutic effect of the observation group was better than that of the control group.(2)Subcutaneous fat thickness:After treatment,there was no significant difference in frontal subcutaneous fat thickness between the two groups.After 3 months of treatment,the fat thickness of the two groups increased.Before treatment,the temporal subcutaneous fat thickness of the control group was thicker than that of the observation group.After treatment,there was no significant difference in temporal subcutaneous fat thickness between the two groups.Degrees were higher than those before treatment.(3)Fat retention rate:The survival rate of adipose tissue in the observation group was significantly higher than that in the control group,with statistical significance(P<0.05).(4)Facial improvement:make the facial contour more symmetrical and coordinated,3 months after surgery,the patient’s facial skin has been improved to some extent.(5)VISIA score of color spots in the front and back of the two groups before and after treatment.The VISIA scores of stains,pore and wrinkles in the two groups were improved 3 months after treatment(P<0.05).(6)Complications:In the observation group,1 patient was conscious of facial discomfort 3 months after operation.After examination,no special pathological changes were found.After local massage and hot compress,the treatment was satisfactory.(7)The satisfaction of patients in the observation group was significantly higher than that in the control group,and the difference was statistically significant(P<0.05).Conclusions:Platelet-rich plasma(PRP)combined with fat transplantation is well tolerated.PRP combined with autologous cells can improve facial aging and skin condition to some extent.
