| Objective: To study the effect of autologous adipose-derived vascular matrix component(SVF)combined with autologous platelet-rich plasma(PRP)on the growth of rabbit supraspinatus tendon cells.Methods: The tendon of rabbit supraspinatus muscle was taken under aseptic condition,and the primary tendon cells were subcultured to the fourth generation.Rabbit inguinal adipose tissue was made into autologous SVF,and rabbit central auricular artery blood was made into autologous PRP.The fourth generation tendon cells were divided into four groups.The intervention measures were PBS buffer(blank control group),autologous PRP,autologous SVF,autologous SVF with autologous PRP.The proliferation of cells in each group was measured by CCK counting method,the migration ability of cells in each group was detected by cell scratch test,and the secretion of MMP-2 by cells in each group was measured by Western Blot.Results:1.CCK-8 results showed that the cell proliferation rate of autologous SVF group,autologous PRP group and autologous SVF with autologous PRP group was significantly higher than that of PBS buffer group at 24 h,48 h and 72 h,and the proliferation rate of autologous SVF with autologous PRP group was higher than that of autologous SVF group and autologous PRP group.2.The results of cell scratch test showed that there was no obvious migration in all groups at8 h,but there was obvious migration of autologous PRP with autologous SVF at 22 h.At this time,there was a little migration in autologous PRP group and autologous SVF group,but no migration in group A.At 30 hours,there was obvious migration in group B,group C and group D,but no obvious migration was found in group A.At 46 h,all the cells in the four groups were full of scratches.3.Western Blot results showed that the expression of MMP-2 was the most obvious in autologous SVF with autologous PRP group.The expression of MMP-2 in autologous SVF with autologous PRP group,autologous SVF group and autologous PRP group was significantly higher than that in PBS buffer group(P<0.05).Conclusion: 1.Autologous SVF can promote the proliferation of individual number of rabbit supraspinatus tendon cells and the migration ability of tendon cells and promote the ability of tendon cells to secrete extracellular matrix;2.The combination of autologous SVF and autologous PRP promotes the proliferation of the individual number of rabbit supraspinatus tenocytes,the migration ability of tenocytes,and the secretion of extracellular matrix by tenocytes. |
