Objective: To study the effect of Huazhuo capsule on the diversity and structure of intestinal flora in obese rats induced by high fat diet,and to explore the mechanism of traditional Chinese medicine in the treatment of metabolic diseases caused by obesity.Methods: Sixty-six SPF Wistar male rats(200 to 20 g,60 days of age)were selected,and12 were randomly selected as blank group(KB)and 48 as model group(MX)(hereinafter referred to as the obesity model group).The model group was fed with high fat diet and then randomly divided into model group(MX),metformin group(EJ),Huazhuo capsule high dose group(HG)and Huazhuo capsule low dose group(HD),after successful establishment of the model.All groups were fed the same diet after grouping.The feces of rats were collected at 0,14 and 28 days after drug intervention.The fecal genomic DNA,was extracted for PCR amplification and the Miseq library was prepared,the diversity and structural changes of intestinal flora were analyzed by Illumina Miseq high-throughput sequencing technique.At the end of the experiment,the rats were weighed and the serum triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),high-density Lipoprotein cholesterol(HDL-C),C-reactive protein(CRP)and interleukin-6(IL-6)Were measured.Tumor necrosis factor(TNF-α)and other related indexes.Results:1.Weight change: After 28 days of drug intervention,the weight of rats in EJ,HG,HD group was significantly lower than that in MX group(P<0.05),the difference between EJ group and HG group was statistically significant(P<0.05).2.Blood lipid four items: Compared with the KB group,the levels of TG,TC and LDL-C in the MX group were significantly increased(P<0.01);After drug intervention,compared with MX group,the level of TG,LDL-C,HDL-C in EJ group had better therapeutic effect,and there was no significant difference(P<0.05),in the level of TG,LDL-C,HDL-C(P<0.05),there was no significant decrease in TC level(P>0.05).Serum TG,TC,and LDL-C levels of rats in the HG group all decreased,and the difference was statistically significant(P<0.05),the level of HDL-C was not significantly decreased(P>0.05).The therapeutic effect of HG group was similar to that of EJ group(P >0.05).3.Inflammatory index: The level of CRP,IL-6,TNF-α in MX group was significantly higher than those in KB group(P<0.01).After drug intervention,Compared with MX group,serum CRP,IL-6,TNF-α levels in EJ and HG groups were decreased,and the difference was statistically significant(P<0.05,P<0.01).The CRP of rats in the HD group was decreased,and the difference was statistically significant(P<0.05),the level of IL-6,TNF-α was not signifcan-tly decreased(P>0.05).The therapeutic effect of HG group was similar to that of EJ group(P>0.05).4.The results of intestinal flora analysis showed that:At the gate level,Compared with KB group,the abundance of thick-walled fungi in MX was significantly increased,while the abundance of Bacteromycetes was significantly decreased.After drug intervention,compared with KB group,the abundance of thick-walled bacteria in MX group showed a rising trend,while that in MX group showed a decreasing trend.Compared with MX group,the abundance of thick-walled bacteria in EJ,HG,HD group showed a decreasing trend,while the abundance of Bacteroides in EJ,HG,HD group showed an increasing trend,especially in HG group.At the genus level,compared with KB group,in MX group,the abundance of bacteroides,Spirulina,Lactobacillus,Roeberiella,Rumen cocci decreased,while Prevos increased relative abundance;After 14 days and 28 days of drug intervention,compared with KB group,the abundance of Lactobacillus,Spirulella,Lactobacillus,rosberellae,Rumen cocci showed a dec-reasing trend in MX group,while the abundance of Prevauus showed an upward trend,and that of Helicobacter spp.was higher in the MX group than in the control group.E.coli abunda-nce increased;Compared with MX group,in EJ,HG,HD group,the abundance of Bacteroides,Spirulina,Lactobacillus,Rossellae and Rumen cocci showed an increasing trend,while Prevos had no significant change in EJ and HD groups.In the HG group,there was a significant downward trend.In addition,the abundance of Akkermansia in the genus Vibriobu-tyrate of HG group increased.Conclusion:1.Huazhuo capsule can improve the abundance and diversity of intestinal flora.The results showed that the abundance of thick-walled fungi decreased continuously,and the abundance of mycorrhizal fungi increased,especially in HG group.The abundance of beneficial microorganisms,such as Bacteroides,Spirulina,Lactobacillus,Rosberelia,Rumen cocci,Vibrio butyrate,Akkermansia and so on,increased,while that of the conditioned pathogenic bacteria,Prevos,decreased.2.Huazhuo capsule could decrease the weight of obese rats and decrease the level of serum TG,HDL-C,TC,LDL-C,CRP,TNF-a,IL-6 in obese rats.The mechanism may be related to improving the abundance and diversity of intestinal flora in obese rats.
