OBJECTIVE:To extract Glycol ginsenosides w/o nanoemulsion by ultrasonic alkaline water method,to prepare ginseng saponin w/o nanoemulsion,to screen excellent prescription and process conditions,and to evaluate the anti-aging effect of skin;To prepare ginsenosides anti-aging moisturizers by using nano-milk as raw material,and evaluate the safety and hygiene of moisturizers.METHODS:The single-factor method was used to study the extraction conditions of diol-type ginseng saponins by ultrasonic alkaline water method.The Glycol ginsenosides were purified by column separation and determined by HPLC.The diol-type ginsenosides were used to prepare w/o nanoemulsion,using pseudo-ternary phase diagram to screen the type and amount of nanoemulsion oil phase,emulsifier,co-emulsifier and Km value,type identification,particle size analysis,drug loading,stability,skin accumulation excessive and skin retention were investigated;animal experiments were conducted to investigate the anti-aging effects of nanoemulsions,and hydroxyproline,type I collagen,type III collagen,and matrix metalloproteinase 1(MMP-1),matrix metalloproteinase inhibitor 1(TIMP-1),matrix metalloproteinase inhibitor 2(TIMP-2)content were measured in the skin of each group of mice and statistical analysis;ginsenoside w/o nanoemulsion as raw material to prepare ginsenoside anti-aging moisturizing cream,and it was subjected to skin irritation test,microbial test,pH measurement,viscosity measurement,cold resistance,heat resistance,and centrifugation test.RESULTS:The optimal experimental conditions for the extraction of diol-type ginsenosides by ultrasonic alkaline water were 20℃,pH=13,160 w,20 min,the ratio of material to liquid was 1:10;The best prescription for w/o nanoemulsion was isopropyl palmitate for oil phase,Span 80 was used emulsifier,1,2-propanediol was used co-emulsifier,Km value=2,oil phase and mixed emulsifier ratio was 7.5:2.5;nanoemulsion particle size was 17 nm,the entrapment rate of diol type ginsenoside was 88.5%and the physical stability was good.The cumulative skin permeation and skin retention of the ginsenoside nanoemulsion group were better than the ginsenoside aqueous solution group;the contents of the ginsenoside nanoemulsion high,medium and low dose group of hydroxyproline,type I collagen,type III collagen,matrix metalloproteinase inhibitor 1(TIMP-1)and matrix metalloproteinase inhibitor 2(TIMP-2)were significantly higher than those of blank group and ginsenoside solution group,and matrix metalloproteinase-1(MMP-1)content significantly smaller than the blank group and the aqueous solution group,the skin section showed no pathological lesions;the formula of ginsenoside anti-aging moisturizing cream was determined,the moisturizing cream was non-irritating to the skin,the total amount of microorganisms was less than 10 CFU·g-1,the total number of molds and yeasts was less than 25 CFU·g-1,and no heat-resistant escherichia coli,pseudomonas aeruginosa,golden staphylococcus aureus bacteria was detected.pH,viscosity,cold,heat,and centrifugal stability comply with standards of cosmetics industry.
