| PurposePsoriasis is a common,chronic inflammatory skin disease.Hyperkeratosis and differentiation of keratinocytes,inflammatory cell infiltration,neovascularization are the main pathological changes of psoriasis.Neutrophilic inflammatory infiltration is one of the characteristic pathological manifestations of psoriasis vulgaris.As the body’s first line of defense against the intrusion of microbial pathogens,neutrophils play a very important role in non-specific cellular immunity.Recently,many studies have shown that neutrophils can produce a variety of inflammatory factors involved in immune regulation,playing an important role in the pathogenesis of psoriasis.We previously studied from the cytokine level of Chinese Medicine(TCM)Silver Crumbs No.1 mechanism of treatment of psoriasis.TCM Silver Crumbs No.1 was used to animal models of mice for observation,the experiment showed that it might inhibit the synthesis of IFN-γ and promote the synthesis of IL-4,playing an important role in inhibiting epidermal hyperplasia and improving epidermis and keratosis,thus regulating the balance of cytokine network and participating in anti-inflammation And immune regulation process.However,its mechanism for the secretion of cytokines by neutrophils,and in particular the regulation of Th1 / Th2 balance,remains unclear.Therefore,we will observe the effect of different concentrations of TCM Silver Crumbs No.1 serum on the secretion of IFN-γ and IL-4 by peripheral blood neutrophils in patients with psoriasis vulgaris to explore the relationship between Th1 / Th2 balance and psoriasis In the mechanism of action,thus clarifying the mechanism of TCM Silver Crumbs No.1 in the treatment of psoriasis vulgaris.MethodEighteen patients with psoriasis vulgaris diagnosed clinically and pathologically were collected.Peripheral blood was collected from the patients and neutrophils were isolated and cultured.The purity and activity of the cells were identified by trypan blue exclusion test and then inoculated on the culture plate,4 concentrations(stock solution,1/2 concentration,1/4 concentration and 1/8 concentration)Psoriasis 1,control group plus Tripterygium glycosides tablets,saline,were added above are containing serum,culture 48 After hours,the levels of IFN-γand IL-4 secreted by neutrophils cultured in vitro were detected by ELISA.ResultsIFN-γ: the highest level of IFN-γ expression in the saline group,the lowest expression level in the original solution TCM group.With the decrease of serum concentration of TCM Silver Crumbs No.1,the level of IFN-γ gradually increased.The positive control drug Tripterygium group IFN-γ expression levels higher than the original solution of TCM group,1/2 TCM group.The statistical results showed that there were tatistically significant differences between the original solution TCM group and the saline group(P <0.05),so did it between 1/2 TCM group and the saline group(P <0.05).And there were significant differences between Tripterygium group and the original solution TCM group.(P <0.05)IL-4: 1/4 TCM group had the highest level of IL-4 expression,while the lowest level of IL-4 expression in Chinese herbal medicine group.IL-4expression level increased first and then decreased with the decrease of serum concentration in Chinese medicine.The positive drug control Tripterygium group IL-4 expression level was higher than the original solution TCM group.The statistical results showed that compared with the saline group,1/4 of the Chinese medicine group had statistically significant differences(P <0.05).There were significant differences between the original solution TCM group and Tripterygium group(P <0.05),the 1/2TCM group and the 1/4 TCM group also had significant differences compared with Tripterygium group(P <0.05).conclusionTCM Silver Crumbs No.1 can reduce the expression of IFN-γ in peripheral blood neutrophils in patients with psoriasis vulgaris,and suppress the inflammatory reaction and the proliferation of keratinocytes.On the other hand,the up-regulation of IL-4 expression can inhibit the expression of Th1 cytokine IFN-γ,promote Th1 to Th2 conversion,correct Th1/ Th2 drift,which may be part of the mechanism of TCM Silver Crumbs No.1 treatment of psoriasis. |
