| Objective Observation of acupuncture "Shuigou" on bilateral striate body of rats with ischemic apoplexy extracellular Glutamic acid(glutamic acid and Glu),gamma aminobutyric acid(γ-aminobutyric acid,GABA),lactic acid,lactic acid,LAC)the change of the content.To clear acupuncture "Shuigou" effect on cerebral ischemia lesion area also its hematogenously neurotransmitter and the regulation of energy metabolism disorder.To explore the regulatory effect of acupuncture at "Shuigou" on cerebral ischemic lesions and distant parts of the neurotransmitter metabolic disorders.Method Choose 100 clean and healthy SD rats were randomly divided into 2 groups,,the rats in the two groups of left and right sides of different microdialysis cannula implantation.After surgery,5 rats were randomly selected as control group,and the other rats were used to model the right middle cerebral artery occlusion(Middle Cerebral Artery Occlusion MCAO,)in rats.Using laser Doppler blood flow monitor to monitor the cerebral blood flow and neurological function score(Bederson score)in rats.Fell by 70%±10%and scoring based on blood flow in the brain is greater than or equal to 2 person judged to be made successfully.Die is to be built according to the success of 49 rats micro dialysis casing embedment technique position different were randomly divided into model group(n=16),the contralateral model group(12),acupuncture group 1(12),acupuncture group 2(9).Acupuncture group and acupuncture group 2 insertion probe balance after 1 h,0.25 mm*13mm filiform needle acupuncture Shuigou,thorn into 5mm,the needle for 30 minutes.In 21/min flow rate in rat striatum microdialysates collected during acupuncture by microdialysis,every collection 30min.Continuous acupuncture for three days,daily at the same time to collect the dialysis fluid at the same time,and 7 days after the model is without acupuncture,but to collection of dialysis fluid.The other three groups were collected at the same time and frequency.Using HPLC fluorescence/uv method(High Performance Liquid Chromatography with Fraunhofer Line Discriminator or Ultraviolet,HPLC FLD/DV)outside the striatal cells of rats was detected in Liquid Glu,the dynamic changes of the content of GABA and Lac.Results(1)Glu results show:the model group,the contralateral model group 1,2,3,7 days each time period in the dialysate Glu levels were significantly increased in the blank group(P<0.05),In which the contralateral model Glu content is lower than the model group(P<0.05);Compared with model group,acupuncture group 1’s Glu content in each period were decreased(P<0.05);Each time Glu content also in acupuncture 2 group were lower than that of the contralateral model group(P<0.05).(2)GABA results show that:the model group,the GABA content contralateral model group and blank control group rats at each time in the dialysate increased significantly(P<0.05);and the model group ratio,acupuncture group 1 in the second,third,seventh day between dialysate GABA content was increased(P<0.05);acupuncture group 2 GABA content and the side model group compared to similarly in the second,third,seventh day differences and acupuncture group 2 GABA content higher than on the side of the model group(P<0.05).(3)Lac results show:The model group and the lac side of model group was compared with significant difference(P<0.05),model group rat striatal Microdialysate lac content higher than contralateral model group(P<0.05).The content of Lac in four groups was lower than that in model group(P<0.05),and the content of Lac in the 2 groups was lower than that in model group,which was lower than that in the model group(P<0.05)Conclusions 1.Cerebral ischemia in the hindbrain energy metabolism disorder is not only confined to the lateral ischemic cerebral hemisphere,But present bilateral neurotransmitter metabolic disorders,namely after ischemia brain exist distant dysfunction;2.Acupuncture "Shuigou" acupoint can effectively reduce ischemic stroke rats with bilateral striatum of extracellular fluid in Glu and lac content,increase GABA content,Adjusting the cerebral ischemia caused by whole brain neurotransmitter metabolic disorder. |
