The Experimental Study Of Shugantongluo Decoction’s Influence On IL-6、TNF-α And Substance P In Rat’s Prostate Tissue

Objective To observe the effect of Shugan Tongluo Decoction on experimental rat prostate wet weight,prostatic histomorphology change and effect on the expression of IL-6 and TNF alpha)and substance P(SP)and evaluate the curative effect.And to explore the possible mechanism of treatment by Shugantongluo Decoction,in order to guide the clinical selection of effective traditional Chinese medicine and prescription to provide ideasMethods March age weight of 180-220g male Wistar rats 60,randomly selected 10 as blank control group,the other 50 for the animal model group.The rat model of chronic non bacterial prostatitis was induced by injection of complete adjuvant into the ventral lobe of the prostate,then,The rats were randomly divided into Shugan Tongluo Decoction high dose group,middle dose group,low dose group,positive medicine group,and was given dose according to lml/100g respectively.The sham operation group and the model group were given the same dose of distilled water.All the above groups were given gastric administration,once a day,continuous administration for 30 days.All rats were sacrificed at 24h after the final administration,extraction of prostate tissue,the tissue morphology changes of prostate tissue were observed under light microscope.Finally,the level of IL-6、TNF-α and P substance in prostatic tissue were detected in different groups by means of ELISA.Results ShuganTongluo Decoction has great positive influence in the rats prostate tissue.Compared with the normal group,the levels of P substance、IL-6 and TNF-αsignificantly increased in prostatic tissue of the model group,There are striking differences between the two groups above(P<0.05、P<0.01).Compared with the model group,the levels of P substance and TNF-αobviously decreased in prostatic tissue of each Shugantongluo Decoction group and positive medicine group(P<0.05、P<0.01).Conclusion Shugantongluo Decoction has obvious therapeutical effect on experimental chronic non bacterial prostatitis.Shugantongluo Decoction can obviously improve local inflammation of prostate and regulate local immunity state.Shugantongluo Decoction can regulate the chronic non bacterial prostatitis rat tissues of substance P,TNF-and IL-6.The research demonstrated that the possible mechanism of Shugan Tongluo Decoction in the treatment of CNP is closely related to the influence of adjusting cytokine levels in prostate tissue,reducing the inflammation and protecting the prostate tissue.
