ObjectiveTo study the expression changes and functions of microRNA in the nerve stump after combined electrical stimulation treatment in rats after peripheral nerve injury,and explore the mechanism of electrical stimulation to promote the repair of peripheral nerve injuryMethodsSelect healthy adult male 7-week-old rats to establish electrical stimulation combined with sciatic nerve injury model and sciatic nerve injury model.After 1,7,and 14 days,the total RNA of the sciatic nerve was extracted,and high-throughput sequencing was used to screen the differential microRNA expression after injury;Use bioinformatics software to predict the target gene of the screened microRNA based on the results of high-throughput sequencing,and perform GO and KEGG enrichment analysis,and finally predict its relatively complete signal pathway.ResultsCompared with the control group on the 1 day after surgery,,4up-regulated microRNAs and 13 down-regulated microRNAs were detected in the 1-day experimental group.Among them,rno-miR-6321(50 times)was the most up-regulated,and rno-miR-881-3p(112 times),rno-miR-871-3p(104times)was the most down-regulated;the 7-day experimental group compared with the control group,a total of 14 up-regulated and 12 down-regulated were detected,and the expression was up-regulated The largest is rno-miR-7b(3times),and the most down-regulated is rno-miR-741-3p(95 times);the 14-day experimental group compared with the control group,a total of 8 were detected to be up-regulated and down-regulated There are 10 of them,of which rno-miR-196a-3p(20 times)is the most up-regulated,and rno-miR-702-5p(26 times)is the most down-regulated;compare the 1-day,7-day,and 14-day experiments The miRNA expression level of the group found that there were 9 up-regulated miRNAs and 4 down-regulated miRNAs.The biggest difference in up-regulation was miR-31a-3p,and the biggest difference in down-regulation was miR-490-3p(11 times)and miR-223-3p(38 times).Conclusion1.In the 7-day group,the most obvious difference in expression is miR-7b;in the 14-day group,the most obvious difference in expression is miR-196a-3p.The expression of miR-881-3p and miR-743a-3p,which are consistent and down-regulated in the 7-day group and the 14-day group,may be related to the repair of peripheral nerve injury.2.miR-490-3p may be involved in the repair of nerve damage by acting on the target gene HMGA2.
