| Objective:To explore the clinical value of platelet parameters,platelet activation and apoptosis in the diagnosis and treatment of type 2 diabetes mellitusMethods:1.Randomly collected 60 inpatients who were diagnosed with T2DM instead of being admitted to the Department of Endocrinology of the second people’s Hospital of Lianyungang City from April to September 2020 as the treatment group,selected 60 newly diagnosed and untreated patients with type 2 diabetes mellitus as the primary group,and selected 60 healthy patients who were excluded from T2DM in the same period as the normal control group.The platelet count(PLT),platelet distribution width(PDW),mean platelet volume(MPV)and hematocrit(PCT)of the three groups of subjects were measured by automatic hematology analyzer.AA(arachidonic acid),ADP(adenosine diphosphate)and COL(collagen)were used as inducers to detect the maximum platelet aggregation rate of 300s in three groups.(PAgTm);was used to detect the expression of platelet P-selectin(P-selection),PAC-1,mitochondrial membrane potential(Δψ m)and intracellular ROS in treatment group and normal control group by flow cytometry.Westernblot was used to detect the expression of platelet apoptotic protein cytochrome C,Bax and Casepase3 in treatment group and control group.2.Retrospective analysis of the laboratory examination results of T2DM patients.The urine routine test data of T2DM patients from April to September 2020 were collected by LIS.According to the level of urine glucose,the patients were divided into three groups:"1+,2+,3+".The results of fasting blood glucose,glycosylated serum albumin and glycosylated hemoglobin were measured on the same day in each group.To explore the relationship between urinary glucose and fasting blood glucose,glycosylated serum protein and glycosylated hemoglobin under different urinary glucose levels.The data were analyzed by SPSS17.0 software single factor analysis of variance(One-Way-ANOVA).The quantitative data of different groups were expressed by mean ± standard deviation,and the test results of different groups were compared by S-N-K methodResults:1.The MPV of the treatment group was significantly higher than that of the normal control group,but the expression rates of PCT,PDW,PLT,AA-PAgTm,ADP-PAgTm and COL-PAgTm were not significantly different between the two groups(P>0.05).The expression rates of MPV,AA-PAgTm,ADP-PAgTm and COL-PAgTm in the primary group were significantly higher than those in the normal control group and the treatment group(P<0.05),but there was no significant difference in PCT,PDW and PLT among the three groups.The expression levels of cytochrome C,Bax,Casepase3,P-selectin(P-selection),PAC-1,mitochondrial membrane potential(Δψ m)and intracellular ROS in the treatment group were significantly higher than those in the normal control group.2.There were significant differences in fasting blood glucose and glycosylated serum albumin among different levels of urinary glucose(P<0.05),but there was no significant difference in glycosylated hemoglobin among different groups(P>0.05)Conclusion:Mean platelet volume(MPV),platelet aggregation rate(PAgTm),platelet activation and apoptosis can be used as evaluation indexes of T2DM to assist the diagnosis and treatment of T2DM. |
