Objective: To investigate the expression of stromal cell-derived factor-1(SDF-1)and protein kinase B(AKT)in mouse wounds at different time by Porcine Acellular Dermal Matrix(pADM),which is the basis for studying the mechanism of wound healing.Methods: Eighteen C57BL/6 mice were treated with a spine as the midline,and a fullthickness skin defect with a diameter of 6 mm was made on the left and right sides.The left side was covered with gauze(control group)and the right side was covered with particulate pADM(pADM group).The tissue at the margin was taken at 1,2,3,4,6,and 8 weeks after injury.Immunohistochemical staining was performed to detect the protein expression of SDF-1 and AKT,and the expression of both proteins in histiocytes was observed pathologically.Results : The expression of SDF-1 and AKT proteins in wound tissues of mice was different at 1,2,3,4,6,and 8 weeks after the establishment of the model,and there were significant differences(with F=45.719,94.905;with P=0.000,0.000).The expression of SDF-1 protein in pADM group and control group was different,and there was significant difference between the two groups at the 3rd and 8th week(with t=8.559,-7.042;with P=0.001,0.002).There was no significant difference between the two groups at other time.AKT protein expression was significantly different between the two groups at week 2,3,4,6 and 8(with t = 6.097,8.462,-9.711,-8.296,-6.079;with P = 0.004,0.001,0.001,0.004),but there was no significant difference between the two groups at first week.AKT and SDF-1 had similar trends in pADM group,both of them began to increase in the 2nd week and reached the peak in the 3rd week.At this time,the difference of expression of AKT and SDF-1 was the greatest in pADM group and control group,and it began to decline after the third week and rise again in the eighth week.Immunohistochemistry showed that SDF-1 and AKT were expressed in the wound tissue,and the positive cells were mainly distributed around the hair follicle cells.Conclusion: pADM may promote wound healing or hair follicle repair by promoting the expression of SDF-1/CXCR4 signaling pathway and downstream signaling factor AKT.
