The skin is the largest organ of body.As the surface of the body,the skin is a barrier covered on the body's surface,which plays the role of keeping moisture and maintaining the normal physiological function.When skin is affected by external injury or the factors of disease and damage of the skin come into being,it may lead to a significant risk,even endanger people's lives.The recent research found that cell therapy contributes to the repair of skin lesions.In this paper,we mainly investigated the effects of the sweat gland-like cells which induced by the rat bone marrow mesenchymal stem cells(rBM-MSCs)combined with sheep acellular dermal matrix(ADM)on wound healing.First,the material sources used in this experiment were the bone marrows of rats,and the target cells were obtained by using the whole bone marrow adherence method.Morphology,proliferation and surface markers were used to examine the results,which showed that the obtained cells were rBM-MSCs.In this study,cell induction was conducted by the co-culture of rBM-MSCs and sweat glands cells(SGCs).RT-PCR and cellular immunofluorescence were used to detect the results,and it showed that the cells were sweat glands cells.Subsequently,the sheep ADM was prepared by enzymatic digestion,the cytotoxicity was detected by MTS and the degree of acellular was examined by HE staining.The results showed that the acellular degree of ADM was relatively high and there was no obvious cytotoxicity.Finally,ADM combined with sweat gland-like cells and the one that was uncombined were covered on nude mice injured wounds respectively.By measured the rate of wound healing and the expression of protein(VEGF/Collagen-I)which involved in wound healing and detected the relative expression of apoptosis-related genes(Bax/Bcl-2)in healing wounds.Finally,we concluded that sweat gland-like cells combined with sheep ADM inhibited the apoptosis of wound tissue and accelerated the rate of wound healing.In conclusion,the sweat gland-like cells combined with sheep ADM inhibited the apoptosis of wound tissue and accelerated the rate of wound healing.In this study,we investigated the effects of rat sweat gland-like cells combined with sheep ADM on wound healing,and the aim of this study was to provide the basis for further study on the mechanism of sweat gland like cells combined with sheep ADM in wound healing.
