Background:The Chinese cobra is one of the most venomous snakes in China.Its poisoning can cause swell,pain and necrosis of the skin and soft tissues under the local position.When the venom enters body,it can cause pathological changes in many organs,which can lead to MODS and even MOF,as well as life-threatening.The Chinese cobra toxin contains mainly cytotoxin,phospholipase A2,etc.,the main components are proteins and peptides;the amount of detoxification of a bite is about 79.1 mg.Currently,most research on Chinese cobra focuses on snake venom ingredients and clinical treatment.There is a lack of research on the clinical models of animal poisoning from Chinese cobra bites,the body’s release of toxins and pathogenesis.Therefore,through new technologies and methods,we will establish a scientific animal model accorded with clinical features and research the circulate way by the Chinese cobra toxin in the body.It is vital to formulate a new and effective clinical treatment plan,relieve clinics patients and improve the patient’s prognosis.Objective:The aim of this study was to establish a model of SD rat that corresponds to the clinical features of Chinese cobra poisoning and to investigate the circulation route of the Chinese cobra toxin in the body.Methods:Select the correct lower extremities of adult SD rats,inject different doses of Chinese snake node venom solution,observe and compare their clinical manifestations and survival status,examine the clinical reality of refractive injection test for rat venom infiltration and target compliance with Chinese sting poisoning SD rat model with consistent clinical manifestations and features to determine standardized modeling methods.On this basis,the dry snake venom powder was dissolved and subjected to transpiration and adjusted to p H 8.0 with Na HCO3(PH=9.5),and then bonded to the fluorescent Cy7 dye,reacting at room temperature for 2 hours and denatured.Protein was removed by high-speed centrifugation at 12,000 rpm and separated by a color column.Clean Cy7 to collect Cy7-labeled protein tops to remove free Cy7.Cy7-labeled Chinese cobra toxin was injected into the lower limbs of SD rats at the modeled dose at 0 hours,5 minutes,15 minutes,30 minutes,1 hour,2 hours,4 hours,6hours,12 hours,24 hours,The fluorescence diffusion range was observed with small animal imagers.The heart,liver,spleen,lung,kidney,muscle,and lymph nodes of the SD rats were taken for pathological section at 2 hours,4 hours,6hours,and 12 hours,and the tissue and cell necrosis of the corresponding parts were observed.Compare imaging and pathology results of small animal imaging results.Results:1.The content of 1/9 times the initial liquid group is a mixed solution of0.00078 mg/g of dry powder and 0.0154 ul/g of water as the target dose.The mortality rate after injection in the right lower extremity of SD rats closest to 10%after survival analysis.The results of routine blood tests for white blood cell count and platelet count are more compatible with clinical ones.The results of biochemical blood tests for blood creatinine,creatinine kinase isoenzyme and alanine aminotransferase are more compatible with clinical ones.The results of histopathological examinations are visually visible.Swelling and necrosis of local tissues are visible and the imaging findings of cell necrosis such as the solution and fracture of muscle fibers and the disturbed arrangement of muscle cells can be observed under the microscope,which is relatively close to the results of clinical trials.2.After injecting Chinese cobra toxin into SD rats’thigh muscles,the toxin mainly infiltrates and spreads from the injection site to the surrounding area.It has not been found that the Chinese cobra toxin selectively or agglomerates in other organs or tissues such as centripetal,liver,spleen,lung and kidney.Conclusion:1.Dissolve 0.00080mg/g of dry powder of Chinese Cobra snake venom in0.0154ul/g of pure water to prepare a mixed SD rat model that meets the clinical characteristics of poisoning from Chinese cigarettes.2.The spread of Chinese cobra toxin in the body may be a mixed circulation mode which is dominated by local infiltration and diffusion and co-exists with other circulation modes.
