Objective: To investigate the antitumor effect of the membrane toxin 12(MT-12) from cobra (Naja naja atra) venom on orthotopic transplanted bladder tumor in experimental animal, and preliminarily research whether the MT-12 can induce apoptosis in the transplanted tumor and its mechanism. Methods: Establishing orthotopic transplanted tumor model in 40 Balb/c female nude mice with human bladder cancer cell line BIU-87 (well differentiated papillary human bladder Tumor cell line). TCC cells were grown in vitro through monolayer cell culture. Then every bladder of the 40 Balb/c female nude mice(6 to 8 weeks old)were inoculated with 5×10~6 living bladder cancer cells via open operation. After the bladder was exposed, 125ul suspension of tumor cells were injected to the membrane layer of the bladder by a thin needle. 3 days later, began the intravesicular perfusion with MT-12. These 40 nude mice were randomly divided into 4 groups(n=10):high dose group(1.0mg/kg), middle dose group (0.5mg/kg), low dose group (0.25mg/kg) and control group. In treated groups, the intravesicular perfusion with MT-12 solution of different dose was performed once every 3 days, at the same time, control group were treated by 0.9% NaCl solution.
