The Inner Mongolia Albas white cashmere goat is an excellent goat breed in China,and the high-quality cashmere produced is often used as the material of various textiles.The quality of cashmere is related to the growth and development of hair follicles in the skin tissues of cashmere goats.The cyclical growth process of hair follicles is regulated by a variety of genes and proteins.Studies have shown that the expression of Vimentin is significantly differential in the cell cycle development of the secondary hair follicles of cashmere goats.But its mechanism of action is still unclear.In order to further clarify the mechanism of Vimentin,the promoter sequenceof Vimentin gene is cloned by PCR technology and analysized on bioinformatics,and it is connected with pEGFP-N1 fluorescent vector,transfected into fibroblasts,then activity is detected by immunofluorescence technology.The transcription factor cDNA library of Vimentin is constructed and identification of the interaction protein by yeast hybrid techniques in this experiment.The results showed that:(1)The promoter 2198 bp sequence of Vimentin gene was cloned successfully,and the promoter is dected to have transcriptional activity.(2)The transcription factor cDNA library of Vimentin is preliminary established by yeast hybrid techniques.(3)Vimentin and ACTB genes are cloned successfully and the yeast expression vector is constructed.(4)The interaction between Vimentin and ACTB was preliminarily proved by yeast two-hybrid techniques.
