| [Research background and purpose]Duhuo,is the dried root of Angelica pubescens Maxim.f.biserrata Shan et Yuan.Its main active ingredients are coumarins and volatile oils,which possess various pharmacological activities,such as antibacterial,antioxidant and anti-inflammatory properties.In this study,we investigated the chemical composition of the ethanolic extracts of Duhuo and its antibacterial effect in vitro.The obtained results will help to maximize the exploitation of Duhuo for possible use in nature feed preservatives.[Research methods and results]1.Preparation of Duhuo extract(DE)Duhuo were powdered and sequentially extracted with 95% ethanol by heat-refluxing×2 at 80℃ for 2h.After filtration and evaporation in vacuo,the ethanolic extract of Duhuo.2.Chemical composition analysis1)Qualitative AnalysisThe qualitative analysis of the DE was performed using UHPLC-MS/MS system and GC-MS system.There were 105 compounds,which including 62 new compounds were estimated or identified.Coumarins are important compounds in Duhuo extract,and 29 isomers and 9 new coumarin compounds were detected in DE.2)Quantitative determination of 7 important coumarin compounds in DEThe UHPLC-MS method was successfully used for the simultaneous determination of the 7 analyzed coumarins in positive ion mode.The results indicated that columbianadin and osthole has the higher content in coumarins compounds,were 36.019(mg/g)and 19.022(mg/g),respectively.And followed by isoimperatorin(1.169 mg/g),bergapten(0.722 mg/g),psoralen(0.337 mg/g),imperatorin(0.092 mg/g)and scopoletin(0.068 mg/g),respectively.3.In vitro antibacterial effect of DE in feedFeed samples were randomly divided into four groups,and they are blank group(no addition of antimildew agent),control group(addition of sodium diacetate at 1 g/kg),DE1group(addition of DE at 2 g/kg),and DE 2 group(addition of DE at 4 g/kg).The results showed that DE could effectively inhibit the growth of bacteria and mold in feed.In addition,DE could significantly inhibit the production of ZEA.4.Antioxidant effect of DE in soybean oilFeed samples were randomly divided into six groups,and they are blank group,control group(addition of BHT at 0.02 g/100 m L),DE 1 to 4 groups(addition of DE at 0.1,0.2,0.4and 0.8 g/100 m L,respectively).The results showed that DE could effectively inhibit the oxidation of lipid compounds and the generation of free radicals in soybean oil.[Conclusion]1.Chemical compositionDE contains coumarins,acids,amines,and lactones.And the coumarin compounds are the main type of compounds.2.In vitro antibacterial effect of DEDE could effectively inhibit the growth of bacteria and mold,and significantly inhibit the production of ZEA in the feed.3.In vitro antioxidant effect of DEDE could effectively inhibit the oxidation of lipid compounds and the generation of free radicals in soybean oil. |
