Greater celandine(Chelidonium majus L.),the popular name is wintergreen barberry root.It is a plant genus in the family Papaveraceae.Entire plants can all put in medicine,it has comprehensive pharmacology functions,such as heat-clearing and detoxicating,relieve pain,furunculosis,detumescence, stop coughing and gaspping.In all its chemical compositions,have obviously biological activity and physiological function are isoquinoline alkaloid,such as chelidonine,chelerythrine,sanguinarine and so on, and these alkaloids are all concerned with the antibacterial and antiviral effect.But the research of greater celandine was not enough,nobody extracted total alkaloids and there was no report about the pharmacology of celandine alkaloids until now.On these grounds,this text not only sets up methods of extracting and assaying the greater celandine total alkaloids,but also do the analysis of acute toxicity and chronic toxicity,even more determin the vitro antibacterial effect for the total alkaloids.The obtained main contents are as follows:1.Extracte technology of greater celandine total alkaloidsThe text apply two extraction lines,they are ion-exchange resin method and macroreticular resin method, that compare the yield and contents of total alkaloids,to pick out the best method.The consequence indicates that the yield and contents of total alkaloids from ion-exchange resin method are 3.67%and 51.61%,are more higher than macroreticular resin method,they are 2.74%and 35.54%.So we take the ion-exchange resin method to extracte the greater celandine total alkaloids.2.Acute toxicity experiment of greater celandine total alkaloidUses the improved Karber's method to determin the mouse's LD50.The computation of LD50 is 1222.55mg/kg,and the LD50 95%average credible limit is 1083.93~1377.21mg/kg,so we conclude it is the low toxic matter.The outset dosage(800 mg/kg) that fixed-dose procedure gives the greater celandine total alkaloids to cause the mouse to be poisoned lethal response is very high,which is 1918.6 times approximately as for the acceptable daily most greatly intramuscular injection dose for the adult.According to the Chinese pharmacopoeia(2005 edition),using in the scope recommended,the greater celandine total alkaloids is of safety. 3.Subchronic toxicity testing of greater celandine total alkaloidsAccording to the LD50,compute the administration dose of each groups,to carry out the subchronic toxicity testing,and determin the blood,physiological and biochemical indicators of rat.The experimental result indicates that the total alkaloids have no influence in hematology index,hemopoiesis and cytopoiesis to rat,and also have no damage or loss effects to hemocyte appearance,liver,renal function.4.Antibacterial effect in vitro of greater celandine total alkaloidsUses flat plate double dilution to determin the MIC of colibacillus,staphylococcus aureus,streptococcus, pasteureLLa bacillus and salmonella.The experimental result indicates that greater celandine total alkaloids have the most depressant effect to streptococcus,next are colibacillus and staphylococcus aureus, and they also have some depressant effect to pasteureLLa bacillus and salmonella.The minimal inhibitory concentration(MIC)is 3.2mg/ml,6.4mg/ml,6.4mg/ml,12.8 mg/ml and 12.8 mg/ml.
