Breeding traits have high economic value in goat breeding,and increasing litter size of ewe is of great significance to improve the production efficency of sheep raising industry.The multiple lambs gene of ewe is the key factor to determine the reproductive traits of cashmere goats,while the number of lambs is a very low heritability quantitative trait,and its heritability is only about 0.1.Therefore,it is difficult for traditional breeding techniques to improve the number of lambs.Because the screening of single nucleotide polymorphism sites(SNP)is convenient,the statistical test is accurate and efficient,and the molecular assistant technology is easy to be used in practice and widely used,it is welcomed by many breeders at home and abroad.The purpose of this study was to analyze the network regulation between GDF9 、BMPR-IB 、 FecB and ESR as candidate target genes and to reveal their potential regulatory mechanism during ovarian tissue reproduction in Liaoning cashmere goats.Biological analysis of non-coding RNA of high propagation genes was carried out to explore the effect of non-coding RNA on lambing rate in goat ovary;The relationship between GDF9、BMPR-IB、FecB and ESR gene polymorphism of Liaoning cashmere goat and lambing number and cashmere yield was identified.The potential SNP,was found by sequence alignment and the GDF9、BMPR-IB、FecB and ESR genes of Liaoning cashmere goat were detected.The effects of polymorphism on lambing number and cashmere yield were analyzed.The results were as follows:1.Detection results of non-coding RNA fluorescence quantitative PCR of high candidate genes:The non-coding RNA in ovarian tissue of Liaoning cashmere goat were detected by fluorescence quantitative PCR.The results showed that the expression of 14 mi RNAs was low in the ovary tissue of Liaoning cashmere goat,the expression of twins was relatively high,and the relative expression of mir-34 c third fetus was the lowest.The ceRNA network to regulate the number of lambs was constructed,and the network of circ RNA-mi R34c-ESR and MSTRG.3853.4 control was obtained,which may play a positive / negative role in regulating the number of lambs produced in Liaoning cashmere goats,respectively.2.Analysis of Polymorphism and Reproductive Traits of High-Frequent Gene:In Liaoning cashmere goat population,Polymorphism was detected in the 3’ UTR region of GDF9、BMPR-IB、FecB and ESR genes,A total of 8 single nucleotide polymorphism(SNPs)sites were found,Of these,5 are effective SNPs sites(C47T、C94T、C299T、C463T and T575G).Genotype analysis showed that,The CC and GG genotypes in Liaoning cashmeregoat population were the dominant genotype of double lambs;The combination of CCGG and GGCC haplotypes is the dominant haplotype of double lambs;The CC genotype of FecB C94 T and ESR C463 T loci is the dominant genotype of double lambs,effect on litter size(P<0.01)of Liaoning cashmere goats,and C alleles were positively correlated with the number of lambs;Therefore,FecB C94 T and ESR C463 T are the main SNPs.affecting litter size of Liaoning cashmere goats a comparative analysis of cashmere yield traits revealed that,FecB C94 T locus TT genotype had the highest average pile yield,fine velvet;BMPR-IB C47 T locus CT genotype individuals had the highest natural villi,ESR T575 G locus GT genotype individuals had the highest net pile rate;The CC genotype of C463 T locus had a significant effect(P<0.01)on villi. |
