Pleurotus tuoliensis,as a cultivated edible fungus species with independent intellectual property rights and clear origin in China,is named for its fresh,crisp and smooth taste and white and fat fruiting body.In recent years,Pleurotus tuoliensis is favored by more and more people in the market.Due to the vigorous development of Pleurotus tuoliensis industry and the increase of production capacity,the problems in the production of Pleurotus tuoliensis have attracted much attention.In the process of industrialization,the abnormal growth and slow growth of the bacteria package or rod often occur,and the specific causes need to be studied.Let the growth of Pleurotus tuoliensis grow normally or accelerate,and improve the slow growth of Pleurotus tuoliensis has become the research topic in this paper.Through the research,we find that acetyl-Co A in the cytoplasm of Pleurotus tuoliensis is the key pivotal intermediate in the growth process.The influence of acetyl-Co A content on the growth of Pleurotus tuoliensis and the influence of different exogenous additives on the growth of Pleurotus tuoliensis can solve the problem of Pleurotus tuoliensis growth The results of this paper are as follows:1.The growth rate of Pleurotus tuoliensis mycelium was affected by exogenous addition.The results showed that different exogenous substances had different effects on the growth of Pleurotus tuoliensis mycelium.Compared with the control group(CK),the growth rate of the treatment with 2.5 m M calcium acetate and 0.5 g / L lignin is faster than that of 7.95 mm / d of CK;when 10 m M calcium carbonate was added,the growth rate is 10.59 mm / d,which was obvious.However,when 20 m M calcium acetate was added,the growth rate was significantly decreased and the growth was inhibited.These substances were related to the synthesis of acetyl-Co A.2.The growth of Pleurotus tuoliensis is positively correlated with the content of acetyl-Co A.The increase of acetyl-Co A content could promote the growth of Pleurotus tuoliensis.The content of acetyl-Co A in Pleurotus tuoliensis mycelia is determined by different treatment methods.According to the determination of acetyl-Co A content in the mycelia of Pleurotus tuoliensis,it was found that the growth rate was positively correlated with the content of acetyl-Co A in the mycelia.3.The metabolic pathway of acetyl-Co A in Pleurotus tuoliensis cytoplasm is analyzed.The mycelia of Pleurotus tuoliensis grown under four different exogenous conditions are selected for transcriptome analysis.A total of 3433 differential genes are identified.Then,the functional enrichment analysis of these differential genes showed that the differential genes were mainly concentrated in the oxaloacetate hydrolase gene,glyoxylate metabolic pathway and related protein synthesis.Acetyl-Co A is a dynamic substance,which is affected by many pathways.For example,in pyruvate metabolism pathway,acetyl-Co A synthase and acetyl-Co A hydrolase,D-lactate dehydrogenase(cytochrome),L-lactate dehydrogenase(cytochrome)and other enzymes jointly determine the content of acetyl-Co A,Therefore,the analysis of acetyl-Co A content under different exogenous treatment conditions should be integrated with multiple approaches.
