As one of the effective methods to block the transmission of infectious diseases,disinfectants can kill or remove pathogenic microorganisms in the external environment,thus cutting off the transmission of pathogens.Compared with foreign countries,the environment of farms in China is relatively poor,and there are some problems,such as relatively single type of disinfectants,narrow germicidal spectrum or high irritation of unilateral disinfectants.Therefore,in this study,benzalkonium bromide and decyl methyl bromide in quaternary ammonium salts were combined with glutaraldehyde to develop a broad-spectrum,high-efficient and low-toxic solution-type veterinary foam disinfectant,and the HPLC method was established to determine the content of benzalkonium bromide,and then evaluate the disinfection efficacy and influencing factors,in order to provide effective disinfection products for the prevention of epidemic spread in aquaculture.This study is mainly divided into the following three parts:1.Prescription development:according to the Technical Standard for Disinfection(2002 edition),Bacillus subtilis was used as the indicator bacteria to carry out suspension quantitative germicidal test,and the germicidal efficacy of the tested bacteria under each proportion was compared to determine the best proportion of germicidal drugs.Determine the best prescription through comprehensive screening,and screening excipients.The results showed that the ratio of glutaraldehyde:benzalkonium bromide:decylmethyl bromide was 2:1.5:3.The main components of the prescription are 2%glutaraldehyde,1.5%benzalkonium bromide,3%decylmethyl bromide,supplemented with some alcohols as co-solvents,0.3%sodium D-isoascorbate and 0.25%fatty alcohol polyoxyethylene ether-9 as antioxidants and surfactants respectively,and a mixture of 3%decyl glucoside and 3%glycerol as foam emulsifier.2.Determination of benzalkonium bromide in compound disinfectant:the content of benzalkonium bromide in disinfectant was determined according to Chinese Veterinary Pharmacopoeia(2015 Edition)and Disinfection Technical Specification(2002 Edition).The HPLC method for benzalkonium bromide was established and verified by methodology.The results showed that the method was sensitive,accurate and durable.Finally,the content of benzalkonium bromide in the pilot test samples of three batches of compound quaternary ammonium salt foam disinfectant was 98.51%,99.46%,all within the range of 90%-110%of the labeled amount.3.Evaluation of disinfection efficacy in laboratory:according to the national standard Technical Specification for Disinfection(version 2002)and Technical Specification for Veterinary Disinfection(version 1992),suspension qualitative and quantitative germicidal test methods were used to compare the disinfection efficacy of compound quaternary ammonium salt glutaraldehyde solution and Baidusha disinfectant with this disinfectant.The results showed that the disinfectant was diluted under the effective dilution multiples of 1:9000,1:30000,1:30000 and 1:2.The disinfection efficacy of Escherichia coli 8099,Staphylococcus aureus ATCC6538,hemolytic streptococcus B 32210 and Bacillus subtilis 63501 treated with 1min,5min,5min and 60min were all qualified.The results of virus killing test showed that the killing rate was 100%when diluted for 3min which was similar to the disinfection effect of compound quaternary ammonium salt glutaraldehyde solution on bacterial vegetative forms,bacterial spores and classical swine fever virus Thiveral strain.The results of influencing factors and article damage test show that temperature can effectively improve the disinfection effect of Bacillus subtilis 63501.This disinfectant is slightly corrosive to copper,non-corrosive to other metals,bleaching to textiles and slightly colored to latex products,almost no damage,indicating that the disinfectant has good disinfection performance.To sum up,the compound quaternary ammonium foam disinfectant developed in this study has the advantages of simple use,rich foam and good disinfection effect,and is suitable for further clinical evaluation and demonstration in animal husbandry. |
