Study On Safety Evaluation、Pharmacodynamics And Pharmacokinetics Of Meloxicam Oral Solution

The pharmacokinetics and pharmacodynamics of meloxicam were studied in this paper.1. The safety evaluation of meloxicam oral solution in dogs and catsTwenty-four healthy dogs and twenty-four healthy cats were allocated in four groups randomly of6samples in each group with balanced gender evenly. Animals in the differernt group were given the dose of meloxicam Omg(control), recommended dose, three times of recommended dose, five times of recommended dose as the per body weight individly by oral adminnistration for5days continuously. Its safety is evaluated by means of general clinical observation, blood routine, indexes of blood biochemical inspection, pathological anatomy and histological observation. The results suggested that:all indexes inspected had no significant difference among the four groups.2. The pharmacokinetics of meloxicam oral solution in dogs and catsMeloxicam was applicated to eight dogs and eight cats at the recommended dose. Blood samples were collected at different intervals and the Meloxicam concentrations in plasma were detected by RP-HPLC system. The plasma concentration versus time datas of Meloxicam in both dogs and cats were adequately described by a one-compartment open model. The Mean±SD, value of T1/2ka, T1/2ke, Tpeak, Cmax, AUC, CL/F, V/F in dogs were0.95±0.64h,23.11±5.99h,4.32±2.15h,0.77±0.25μg·mL-1,28.73±11.20mg·h-1mL-1,0.01±0.003L·h-1,0.25±0.09L·kg-1, respectively; wheras the respective value in cats were0.75±0.45h,6.24±2.21h,2.36±0.95h,0.30±0.11μg·mL-1,3.57±1.49mg·h-1mL-1,0.03±0.02L·h-1,0.27±0.11L·kg-1. The results indicated that the Meloxicam could be easily absorpted in both dogs and cats, the elimination of the drug tend to be slow, it maybe perform a better effects.3. Pharmacodynamics studies of meloxicam oral solutionExperiments of fever induced by peptone in rabbits and twisty response induced by acetic in mice were performed. The results showed that Meloxicam had obviously antipyretic and analgesic effects. Compared with control group, the three dosage groups of Meloxicam displayed significantly antipyretic effects on fever induced by potone in rabbits3hours after the treatment(p<0.05). Compared with control group, the three doseage groups of Meloxicam could significantly inhibit twisty response in mice induced by acetic acid (p<0.01),and the inhibitory rat of the high-dose group and the middle-dose group reached more than80%.
