Pathogen Detection And Molecular Identification Of Potato Virus Disease In Fuqing City Of Fujian Province

Fuqing is one of the main potato growing regions in Fujian Province.In the recent years,potato in Fuqing region has suffer from more and more serious virus disease,which has affected the quality of potato and caused enormous economic loss in potato production.the comprehensive and systematic investigation about the main virus species of potato is seldon reported in Fuqing.The purpose of this study is to investigate the occurrence of potato virus disease in Fuqing,and identify the main diseases and pathogen of potato in this region.In 2017,a total of 56 susceptible potato plant samples were collected from fujian fuqing city.The total RNA was extracted from potato leaves by Trizol method.The specific primers were designed and synthesized using the gene sequence of potato related viral shell proteins reported in the literature.Test and identification of potato samples in the region were tested by RT-PCR.PVY,PMTV,PLRV,PVS and quarantine virus PSTVd were detected.The detection rates were 46.43%,16.07%,14.29%,8.93%and 3.57%.The complex infection phenomenon showed the potato samples in this area can be up to 3 kinds of virus infection.Five viruses were detected in 56 potato samples collected in fuqing area of fujian province.The main types of virus were PVY,Through the cloning and sequence analysis of the target band for virus amplification,The homology analysis shows:There were significant differences in CP nucleotide sequence of PVY isolates in fuqing city,the homology rate is 88.2%～99.0%,and the homology of F34 and F27 is 88.2%.By constructing the NJ phylogenetic tree for PVY isolates,there are two major evolutionary branches of PVY isolates in different regions.The PVY isolates F34 and Y4 in this area are clustered with the PVY isolates in Dalian,Germany,Henan.the separation of F27 and the PVY isolates of Beijing is another branch of evolution.There were differences in PSTVd isolates and PSTVd virulent strain(U23058),attenuated strain(M14814)and intermediate strain(AY492084)genes,And only 2 base differences from the attenuated strain.To investigate the virus pathogens in potato in Fuqing District through molecular biology techniques to understand the occurrence and variation of the pathogens.The paper provides a basis for the introduction of potato seed potato in fuqing area and the prevention and control of virus disease.
