American ginseng is a valuable medicinal material with nourishing effects.It contains functional ingredients such as amino acids,volatile oils,polysaccharides and saponins.As one of the main functional components of American ginseng,American ginseng polysaccharide has a variety of biological activities such as enhancing immunity,anti-tumor,hypoglycemic.This study determined the optimal conditions for the extraction of polysaccharide from American ginseng residue components,the physical and chemical properties of polysaccharides of American ginseng were preliminarily studied,and the product of American ginseng polysaccharide oral liquid was developed.This study determined the optimal formulation of the oral liquid and carried out a factory design.The main experiment results were as follows:1.The results of single factor and orthogonal experiments show that the ratio of material to water,the extraction temperature and time were the main factors that affect the extraction rate of polysaccharide.The optimum parameters were as follows:the ratio of material to water is1:20,extraction temperature is 90℃,extraction time is 150 min.2.The analysis of physical and chemical properties show that the critical concentration of American ginseng polysaccharide solution is 1.20 g/d L,and the intrinsic viscosity is 0.96d L/g.American ginseng polysaccharide solution has strong elasticity,and its elasticity is greater than viscosity.In the experiment of scavenging DPPH free radicals,the IC50of American ginseng polysaccharide was 1.6466 mg/m L,indicating that American ginseng polysaccharide has a certain antioxidant effect.The results of hypoglycemic experiment in vitro showed that the inhibition of American ginseng polysaccharide onα-glucosidase andα-amylase is dose-dependent in the concentration range of 1~10 mg/m L,and the higher the concentration of polysaccharide,the higher the inhibition rate.3.Through single factor experiment and orthogonal experiment,the best formula of American ginseng polysaccharide oral liquid is that the concentration of American ginseng polysaccharide is 15 mg/m L,the addition amount of citric acid is 0.15%,and the addition amount of stevioside is 0.06%.The clarification effects of chitosan,centrifugation and static sedimentation were compared,and 0.75%chitosan was determined as the best clarification method.The sterilization effects of adding preservatives and different temperatures and times were compared,and the sterilization process was determined to keep oral liquid at 85°C for15 min.4.The results of stability experiment showed that the polysaccharide content of American ginseng polysaccharide oral liquid had no obvious change within a certain period of time,and the microbiological test met the relevant standards,which proved that American ginseng polysaccharide oral liquid had good stability.In the quality standard of American ginseng polysaccharide oral liquid,the relative density of oral liquid≥1.006,the p H range is 4.0~4.5,the polysaccharide content≥15 mg/m L,and the microbial indicators meet the national standards of health food.5.The site of the American ginseng polysaccharide oral liquid factory with an annual output of 10 million was selected,the technological process and operating points in the production of the factory were established,the material balance,water and electricity balance and equipment selection were carried out according to the production requirements.Developed environmental protection measures for factory production and analyzed the economic benefits of the oral liquid factory,designed the layout of the oral liquid factory and workshop layout,and formulated the technical standards of the American ginseng polysaccharide oral liquid factory.
