problems in the production process of qilong capsule and astragalus formula granules,such as the underutilization of the effective components of astragalus,and the production efficiency cannot be maximized,which indicates that the extraction process needs to be optimizedIn order to optimize the extraction process of Astragalus membranaceus,which is the main raw material of Qilong capsule and Astragalus granules,this paper takes the extraction process of Astragalus membranaceus granules and the extraction process of Astragalus membranaceus in Qilong capsule as a reference to determine the different extraction process conditions.The key aspects of the extraction process of Astragalus membranaceus were comprehensively evaluated from the two aspects of chemical composition and pharmacodynamics.The details are as follows:(1)To investigate the paste yield and effective component transfer rate of astragalus extract and alcohol extract under different extraction conditionsAfter analyzing the production process of the Snapdragon capsules under the guidance of traditional Chinese medicine theory and the formulation granules produced by modern technology,the extraction time and extraction process times of water extraction and alcohol extraction in the key steps of the extraction process of Astragalus membranaceus The amount of solvent extracted was used for gradient setting and optimization.The extraction rate of water extract and alcohol extraction of Astragalus membranaceus under different process conditions was investigated.The method of determining the content of Astragaloside Ⅳ in Astragalus membranaceus Ⅰ.by UPLC-MS/MS method was investigated and its content was determined.The HPLC method was used to determine the aqueous extract of Astragalus membranaceus.The method of determining the content of isoflavone glycosides and determining its content was carried out.The method of determining the content of astragaloside Ⅳ in the extract of Astragalus by HPLC-MS/MS method was investigated and its content was determined.The transfer rate is calculated based on these measured contents and the orthogonal results and variance analysis results are obtained,and the optimum water extraction and alcohol extraction processes are preferred.The experiment proves that from the aspect of the extraction rate and the transfer rate of the active ingredient,the optimal process of water extraction is 8 times of water addition,2 times of extraction,1 hour each time;the best process of alcohol extraction is 10 times of alcohol addition,3 times of extraction The effect of ethanol dosage on the transfer rate of astragaloside was statistically different(P<0.05)(2)To study the effects of total saponins of astragalus on internode vascular growth of zebrafish under different extraction conditionsCombined with the chemical composition and the extraction process of Astragalus membranaceus granules and the extraction process of Astragalus membranaceus in Qilong Capsule,the total saponins of Astragalus membranaceus were obtained by alcohol extraction,and the total saponins of Astragalus membranaceus were used as the research object,and the zebrafish internode blood vessels were applied.Intersegmental vessels,ISV)The symptoms of myocardial cell injury or necrosis in patients with injury were studied.The effects of total saponins of Astragalus membranaceus on angiogenesis of zebrafish were determined,and the rate of angiogenesis was determined.A sensitive,feasible and reproducible saponin of scutellaria saponins was select the best one.The following suggestions are proposed for improving the extraction process of astragalus formula granules:In the alcohol extraction process,the extraction time should be increased appropriately,and the extraction time should be controlled in about 3 hours.In the water extraction process,it should reduce the extraction frequency and select once as optimum times.In addition,for different diseases,the therapeutic substances in astragalus may not be the same,so that the extraction process of astragalus formula granules should be adjusted according to different syndromes and
