Studies On Extraction And Purification Of Astragalus Formula Particles

Astragalus is the dry root of Astragalus membranaceus(Fisch.) Bge.var. mongholicus(Bge.) Hsiao and Astragalus membranaceus(Fisch.) Bge.The main ingredient of astragalus are astragalussaponins and astragalus polysaccharide,research shows that they are of important pharmacological effects.Herbal formula particles as to replace traditional Chinese medicine yinpian,it has convenient,quick,safe,health,science,etc.studies on extraction of herbal formula particles is important,it can improve the efficiency of using resources of Chinese herbal formula,even improve the efficacy of Chinese herbal.We established the analysis method of astragalus saponins and astragalus polysaccharide,optimized the extraction process of astragalus formula particles,study on separation and purification of astragalus extract with macroporous adsorption resin,the results are as follows:1.established the analysis method of astragalus saponins and astragalus polysaccharide,Astragalus medicinal quality inspection.We established vanilla aldehyde - high chlorine acid colorimetric method and anthraquinone - sulfuric acid colorimetric method to determined the content of saponins and polysaccharide in astragalus extract,results show that the method is stable,reliable and accurate.Established TLC scanning method to determined theastragalus nucleoside armour in astragalus, according to the content of pharmacopoeia,check the quality of astragalus, the result shows that the astragalus used in experiment is good with the standard of pharmacopoeia of astragalus.2.study on extraction process of astragalus formula particlesOptimized the extraction process of astragalus formula particles.Through the orthogonal experiment and Single-factor test of Solvent,the factors of the optimum extraction process:first use 10 times the amount,80 percent ethanol extraction for 2 hours,with 10 times of medicinal residue water extraction 2 times,2 hours.3.study on purification process of astragalus extract with macroporous resinThe absorption and desorption test are used to select the better resin.The result shows that the adsorption effect are different with different types acroporous resin.The AB-8 resin is the suitable material.On the basis of the above,with the content of astragalus saponins as index, the kinesis adsorb and elution process with quantity of astragalus saponins are investigated,the maximum quantity and density of physic liquor and the type and quantity of elution solvent are determined,the results show that the,and the elution solvent determined is four times column volume of 80% alcohol,...
