Objective In this paper,Lindera glauca was used as the raw material,through the combination of hot bleaching and toner,solve the process of lindera glauca processing,the optimal extraction process,determine the difference of the antioxidant activity of polyphenols before and after color protection,based on metabolomics technology qualitative and quantitative analysis,the differential metabolites and clear the metabolic process and change mechanism,the influence of color protection process on the metabolism of lindera glauca was predicted and analyzed.Methods Using sodium hydroxide and zinc acetate ironing and bleaching color protection,with the color difference value ?E as the index,the sodium hydroxide concentration,ironing time,ironing drift temperature,and zinc acetate concentration were optimized in combination with the response surfacethe,free radical clearance and total reduction capacity of various concentrations of polyphenols,DPPH and ABTS were determined by comparing with Vc.Analysising the influence of color protection process for lindera glauca polyphenol metabolites by UPLC-MS/MS metabolomics platform and statistical method.Results(1)Through single factor and response surface experiments,the results show that the best color protection condition is 90℃,sodium hydroxide solution concentration0.02%,ironing time 180 s,zinc acetate solution concentration 0.05%,the color difference value is 4.15,the color protection effect is excellent.(2)Comparing the effects of 50% acetone,50% ethanol,methanol-acetone-water(1:1:2 v/v/v)and 50%methanol on the extraction amount of lindera glauca polyphenols,the results showed that the ethanol solution extracted the polyphenols at 31 mg/g,Significantly higher than that in other solvents;The effect of liquid ratio,ethanol volume fraction,time and temperature on polyphenol extraction was investigated by single variate and response surface,The optimal conditions were liquid material ratio of 50:1(m L/g),ethanol volume fraction is 70%,extraction time of 90 minutes and extraction temperature of 80℃,The extraction amount of downhill lindera glauca polyphenols was 31.46 mg/g.(3)Large-porous resin was eluted by an ethanol solution gradient,the elution components after color protection were obtaineds 1-1(20% ethanol)and1-2(40% ethanol),Unprotected color eluted components were obtaineds 2-1(20%ethanol)and 2-2(40% ethanol),lindera glauca polyphenols(1-1,1-2,2-1,2-2),had in vitro antioxidant activity,and the antioxidant capacity of unprotective polyphenols was higher than that of unprotective polyphenols.(4)Through metabolomics identification and analyzing the differential metabolites and metabolic pathways of polyphenols(1-1,1-2,1-2,2-2,2-2),292 differential metabolites were selected in 1-1and 1-2,388 differential metabolites were selected in 2-1 and 2-2,Parsing metabolic pathways,mainly secondary metabolism,biosynthesis,flavonoid biosynthesis,flavonoids biosynthesis,phenylalanine metabolism,tyrosine metabolism and anthocyanin biotin synthesis.Conclusion A technology of lindera glauca color protection was established in this experiment.The extraction process of polyphenols in lindera glauca was established.Qualitative and quantitative analysis of polyphenols in lindera glauca based on metabolomics,visualization of the influence of color protection process on lindera glauca polyphenols metabolites,to provide theoretical support for the study of lindera glauca polyphenols.
