| China’s Marine resources are very rich.Marine collagen polypeptides have biodegradability,high safety and rich biological functions and nutritional value,so they have a wide application prospect.At present,there are few studies on Marine fish resources,and the utilization rate is low.The fish skin and other by-products in Marine fishery are discarded in large quantities,causing resource loss and environmental pollution.Therefore,it is of great significance to accelerate the research,development and application of Marine fishery by-products.Studies have shown that shark skin is rich in collagen.In this study,the skin of the great blue shark was used as the research material to extract collagen polypeptide from the skin for the related efficacy study,laying a theoretical foundation for its development as a functional product,and at the same time providing a certain guiding significance for solving the waste of shark byproducts.Taking the blue shark skin as the research material,the extracted white shark skin polypeptide freeze-dried powder SHP was found to have a molecular weight of less than 1000 Da,a protein content of up to 86.82%,a fat content of only 0.40%,and a heavy metal content lower than the national standard.In the amino acid composition,glycine content is the largest,which is 127.15g/kg.At the same time,it has more proline and hydroxyproline,which has the characteristics of collagen.The ultraviolet full wavelength absorption spectra showed that SHP had the characteristics of polypeptide.It has a certain hygroscopicity,strong moisture retention,and is not easy to lose moisture.To investigate the effect of SHP on HaCaT cells(immortalized human epidermal keratinocytes),MTT assay results showed that SHP could increase the activity of HaCaT cells up to 182.6%.Colony formation experiments showed that SHP promoted the colony formation of HaCaT cells.Cell scratch experiment showed that SHP could improve the ability of cell scratch healing.In terms of antioxidant activity,in vitro antioxidant results showed that SHP could scavenging DPPH·,ABTS+·,·OH and O2free radicals,and the scavenging ability was positively correlated with the concentration.The scavenging effect of·OH radical was the best,with IC50=0.35 mg/mL.The results of cell antioxidant showed that SHP could reduce the intracellular ROS content of L02,scavenging intracellular free radicals,reduce the content of MDA.improve the activity of antioxidant enzyme SOD,and protect cells from the damage of exogenous oxide H202.In the aspect of anti-fatigue activity,SHP can improve the loading swimming time of mice,increase the grasping power of mice,reduce the contents of lactic acid and urea nitrogen in mice serum,and increase the content of serum glucose.In terms of bone strengthening activity,the results of scanning electron microscopy and HE staining of bone sections showed that the parenchymal bone tissue in the bone marrow cavity increased,the area of bone density increased,and the number of bone cells decreased in the SHP group.Subacute toxicity test in mice,body weight measurement,visceral organ removal,visceral organ coefficient calculation and visceral HE staining showed no toxic and side effects,indicating that SHP is safe and non-toxic.This study laid a theoretical foundation for the development of SHP as protein nutrition,wound healing drugs,anti-fatigue,anti-oxidation and bone strengthening products. |
