In recent years,with the changes of people's living standards and lifestyles,the proportion of high-fat and high-calorie foods in daily diet has increased.And the incidence of obesity,"three highs"and cardiovascular and cerebrovascular diseases has increased year by year,which has become one of the public health problems to be solved urgently.As the main active substance of coix seed,Coix seed oil is widely used in anti-cancer and anti-tumor.However,with the upgrading and optimization of extraction technology of coix seed oil,it has certain economic and social benefits as a dietary supplement for daily weight loss and lipid reduction.In this study,,ICR healthy mice were taken as the research object and based on the purification of coix seed oil,the physiological and biochemical indexes were detected through three groups of experiments:short-term high-dose coix seed oil exposed healthy mice,long-term low-dose exposed healthy mice and long-term low-dose exposed high-fat mice,and the effects of coix seed oil on lipid-lowering and weight-reducing of mice were confirmed;At the same time,in order to alleviate the easy oxidation characteristics of coix seed oil,coix seed oil microcapsules were prepared by micro-emulsification with two wall materials,and the stability and in vivo release mechanism of coix seed oil microcapsules were explored.The results are as follows:1.The experimental results of 15-day short-term high-dose exposure and 12-week long-term low-dose exposure to healthy mice showed that the lipid-lowering and weight-reducing effects of coix seed oil were closely related to the dosage and action time.In the long-term experiment,the weight of mice in the low-dose group of coix seed oil decreased by 17.0%,10.79%and 6.80%,and compared with the high-dose group of coix seed oil,it also decreased by 7.64%,while in the short-term experiment,the weight of mice in each group decreased.Long-term low-dose consumption of coix seed oil can effectively reduce the concentration of triglyceride(TG),low density lipoprotein(LDL-C)and TC/HDL-C in serum,but the effect is not significant in short-term experiment.Similarly,coix seed oil can significantly reduce the concentration of aspartate aminotransferase(AST)and alanine aminotransferase(ALT)in serum and effectively improve the level of liver fat,the oxidative stress indexes of tumor necrosis factor(TNF-?)and interleukin-1?(IL-1?)in both groups were in normal range,the liver tissue slices of mice fed with coix seed oil for a long time had no obvious damage,but the liver slices of mice fed with too much oil showed some damage.16S rDNA sequencing analysis showed that the abundance of dominant bacteria in the intestinal tract of mice was changed.2.Coix seed oil can reduce the weight of obese mice,regulate blood lipid and protect liver function.Compared with the high-fat recovery group,the weight of mice in coix seed oil group decreased by 27.3%and 37.3%respectively,which could significantly reduce the serum TC/HDL-C,AST and ALT levels of obese mice.The levels of inflammatory factors TNF-?and IL-1?were no different or lower than those of the normal control group,and the steatosis of liver tissue sections was improved.3.The microcapsule particles formed by soybean lecithin(Soy)are smaller and more uniform,and the system is more stable,the surface of starch octenyl succinate(OSA)microcapsules have depressions,wrinkles and mutual adhesion.The critical micelle concentration(CMC)of Soy and OSA measured by fluorescence spectrometry is 4×10-4 mol/L.The addition of coix seed oil has little effect on the particle size and potential of Soy and OSA emulsions,but the average particle size of Soy emulsion(453 nm)is smaller than that of OSA(608 nm),and the absolute potential of Soy emulsion(61 mV)is higher than that of OSA(42 mV).Under microscopic observation,the morphology of Soy emulsion particles is more complete,and OSA particles are sunken and damaged.With the increase of the addition of the two wall materials,the permeability to the inner and outer membranes of bacterial cells increased slowly.
