At present,cancer has become an important threat to human health.Traditional therapies can damage normal human tissues or reduce the quality of human life.Photo-therapy based on light irradiation has attracted wide attention because of its minimally invasive,accurate and selective nature.Single photothermal therapy or photodynamic therapy has been widely used in cancer research.However,synergistic therapy has at-tracted much attention because its therapeutic effect is higher than that of single photo-thermal therapy or photodynamic therapy.Based on this,a 4-carboxyphenylporphyrin-silica coated gold nanorod(AuNR@Si O2-TCPP)nanocomposite was constructed for the combination of photothermal and photodynamic therapy of tumor in vivo and in vitro.The size of AuNR@SiO2-TCPP nanocomposite prepared in this work is about 190nm.The LSPR peak of gold nanorods is 800 nm in position and the size of gold nano-rods is about 52.8 nm in length and 13.5 nm in width.The optimum operating condi-tions for the preparation of gold nanorods with LSPR peak at 800 nm are aging seed time of 2 h,seed amount of 60?L,HCl of 2.4 umol/L,Ag NO3 of 10.0 mmol/L.The thickness of the coated silica shell is about 66 nm.The temperature of the dispersion of0.11 nmol/L AuNR@Si O2-TCPP can rise to 53.6?after 15-minute irradiation with808 nm laser(1.9 W/cm2).AuNR@Si O2-TCPP can effectively produce singlet oxygen under 660 nm laser irradiation.Synergistic therapy of photothermal and photodynamic effects of tumor in vivo and in vitro was studied.In MTT assay,the survival rate of A549 cancer cells treated with AuNR@Si O2-TCPP(100 ug/m L)was only 21%.In an-imal experiments,A549 tumor growth of mice injected with AuNR@Si O2-TCPP(5mg/kg)and irradiated with(808+660)nm was significantly inhibited,with an average relative volume of 0.78.In control groups this value increased to 7.2.The H&E staining of the tumor tissue showed significant apoptosis of A549 cells.These results indicate that AuNR@Si O2-TCPP is a potential anti-cancer nanodrug.
