Cicada flower is a kind of medicinal and edible fungus with a long history and high medicinal value.Research in recent years has revealed the huge application potential of cicada flowers for diet and medicinal purposes.However,it has also triggered people's frantic picking.Coupled with the continuous deterioration of the ecological environment,the natural resources of cicada flowers have become increasingly tense and artificial cultivation has become a hot spot in domestic and foreign research.Therefore,it is very necessary to explore the important environmental factors that affect the production of cicada flower fruit bodies.n this study,in order to reveal the complex growth mechanism of cicada flower fruit bodies and explore possible interactions between microbial communities,we used high-throughput sequencing to explore the internal and external microbial community structure of wild cicada flowers,and analyzed the flower fruit bodies have an important impact on the physical and chemical properties of the surrounding soil.In addition,for the subsequent cultivation of artificial cicada flowers,we isolated the key dominant fungal strains that lead to the formation of fruiting bodies,and carried out liquid fermentation and solid fermentation respectively on them,so as to obtain artificial cultures close to wild cicada flowers.In this experiment,the sclerotia,coremia and habitat soil of the cicada flowers in Anji County,Zhejiang Province were selected for high-throughput sequencing to reveal the microbial community structure and compare the differences in the bacterial community of each sample.The results showed that the diversity and abundance of microflora in the soil of cicada and peanut environment were significantly higher than that of coremia and sclerotia.A total of 200 bacterial OTUs were detected in the soil,coremia,and sclerotia.Among them,11 phyla,17 classes,43 orders,57 families,78 genera,and 7 phyla,9 classes,27 orders,38families and 59 genera were detected in the soil.There are 7 phyla,8 classes,25 orders,36families and 57 genera in the sclerotia.Fungi were detected in the soil in 8 phyla,24 classes,44 orders,72 families,85 genera,6 phyla,11 classes,22 orders,36 families and 45 genera in coremia,and 4 phyla,8 classes,12 orders,17 families and 17 genera in sclerotia.Among them,the dominant bacterial genera in the soil were Burkholderia-Caballeronia-Paraburkholderia?Bacillus?Acidibacter?f??Xanthobacteraceae?Unclassified?Candidatus?Solibacter,the dominant fungal genera were Isaria?f??Clavariaceae?Unclassified?Umbelopsis?f??Chaetomiaceae?Unclassified?Mortierella.The dominant bacterial genera in the coremia were Pedobacter?f??Enterobacteriaceae?Unclassified?Pandoraea?Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium?Phyllobacterium?Chitinophaga?Stenotrophomonas?Burkholderia-Caballeronia-Paraburkholderia?Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium?Serratia?Pedobacter?Comamonas?Pandoraea?f??Mitochondria?Unclassified?Chitinophaga,the dominant fungal genera were the Isaria?Apiospora?Cladosporium.The dominant bacterial genera in the sclerotia were Achromobacter?f??Enterobacteriaceae?Unclassified?Stenotrophomonas?Burkholderia-Caballeronia-Paraburkholderia?Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium?Serratia?Pedobacter?Comamonas?Pandoraea?Chitinophaga,the dominant fungal genera were Isaria.The strain AH10-4 isolated and purified from the dominant fungi in wild cicada flowers was identified as Cordyceps cicadae strain.After liquid fermentation culture of AH10-4 and other two strains of Cordyceps cicadae,it was found that the fermentation hyphae of AH10-4 showed obvious advantages in biomass,adenosine and HEA content.Therefore,the adenosine content in the fruiting body of the strain was 1.979±0.115 mg·g-1 and 5.815±0.392mg·g-1 after solid fermentation.Compared with natural cicada flowers,the contents of adenosine and HEA in artificial culture products were significantly higher.This study provides new insight into the formation mechanisms of cicada flower and may contribute to the large-scale cultivation of cicada flowers.
