With the increase of global life expectancy,population aging has become an increasingly prominent social problem,research on healthy aging is becoming more important.The long-living people is an excellent model for studying healthy aging due to their high quality of life cycle.In recent years,with the deepening of the study of intestinal flora,more and more evidence shows that intestinal flora plays an important role in shaping human health.Therefore,the composition,structure and function of intestinal flora of long-lived elderly were analyzed to further develop related probiotics and provide theoretical basis for intestinal flora intervention in healthy aging.In this study,we used the feces of the elderly in Dujiangyan city,Sichuan province as the experimental material,screened,isolated and purified it with mucin selective medium to obtain a single colony,and identified the bacteria by sequencing with 16 S r DNA technology.The primary study on the beneficial properties of the strain was carried out by the longevity experiment of model organisms C.elegans and Drosophila.The main research results are as follows:1.The suitable mucin medium was determined and the fecal microorganisms of long-living people were separated and screened.Finally,A total of 108 colonies were selected,17 strains were isolated and identified by 16 S r DNA,including: Enterobacter,Lactobacillus,Escherichia,Bacteroidetes and Enterococcus,which were divided into13 genera.There are three potential probiotics: Lactobacillus mucosae,Pediococcus acidilactic,and Bacillus cereus.Meanwhile,four potential pathogens were isolated,including Enterococcus gallinarum,Enterococcus avium,Escherichia sp.B1147 and Shigella flexneri.2.A rapid screening model of probiotics was established based on the microbehost interaction model with the life span of C.elegans and Drosophila.The bacteria used in the experiment included four strains isolated from long-living people and one strain of Akkermansia muciniphila.It was found that all strains had no significant effect on the life extension of C.elegans and Drosophila.In summary,intestinal microbes of some long-lived elderly people have been isolated in this study,which enriched the bacteria resources of the long-lived elderly people in this laboratory.Potential probiotics among them still need to be further studied.At the same time,this experiment established a set of basic procedures for the separation of anaerobe from the long-lived elderly,which laid a foundation for the further separation of more long-lived elderly bacterial species resources.The establishment of a microbe-model biological interaction platform will provide references for further validation and screening of potential probiotics.
