Objective: To observe the effect of co-culture of serum exosomes and insulin-resistant adipocytes in DB/DB mice after different exercise intervention,and to explore the possible molecular biological mechanism,in order to provide theoretical basis for exercise intervention of diabetes.Methods: A total of 48 8-week-old male db/db mice were randomly divided into 4 groups with 12 mice in each group,which were diabetic quiet control group,diabetic resistance climbing group,diabetic aerobic running group and diabetic swimming exercise group.Another 12 male MM mice were selected as normal control group.Movement after the intervention groups of mice serum,from extracting secrete body outside,will the proposed secrete body and using dexamethasone intervention model of insulin resistance of fat cell culture,in cell supernatant after 24 hours,the expression of glucose and insulin levels,Western blot test groups the expression of PI3 K,and AKT,PPAR gamma,rt-pcr detection and insulin resistance are closely related to the expression of miR-155.Results: 1.The particle size concentration of serum exosomes in each group met the identification criteria.2.After induction and differentiation of 3T3-L1 cells,dense lipid droplets appeared in the cells,the cell morphology changed from fibrous to round,and the typical "ring ring" shape could be seen inside.The oil red O staining could be stained as orange or deep red,which proved that the cell differentiation was successful.3.After 24 hours of dexamethasone intervention,the glucose level in the cell supernatant increased significantly,indicating the successful establishment of the insulin resistance model.4.Glucose levels were significantly increased after MA,DP,DR and DY intervention(P of MA,DP and DR p < 0.001,DY P< 0.01),DA had no significant change before and after intervention(P>0.05).5.The insulin levels of MA,DR and DY decreased significantly after intervention(P< 0.05),insulin levels of DA and DP decreased,but there was no statistical significance(P>0.05).6.Western blot results of exosome intervention showed that PI3 K was significantly upregulated in all groups after intervention(P< 0.001),the expression of Akt was significantly different from that of the control group except for the DA group(P< 0.001),but the expression level of DR was decreased;The expression of PPAR? in each group increased significantly in MA and DY(P< 0.001),there was also a certain difference in DP(P< 0.01),and the expression level of DR was also lower than that of control group(P< 0.001);7.Mi R-155 significantly increased in DP,DR and DY groups(P < 0.001),MA and DA were not significantly different from the control group without exosome intervention;In db/db mice,the exercise and quiet groups were compared again,and it was found that the miR-155 level in the exercise group was significantly increased compared with that in the quiet group.Conclusion:1.Circulating exosomes produced by different exercise modes can reduce glucose levels and increase the levels of miR-155 and related proteins PI3 K and PPAR?,thus improving insulin resistance of adipocytes.2.Endurance exercise,such as swimming and running platform,has better effect on reducing insulin than resistance climbing,and can be used as exercise intervention to improve insulin resistance.
