The normal rheological state of blood makes the tissue have a stable internal environment.The coagulation,anticoagulation and fibrinolysis systems of the human body are dynamic equilibrium process.The occurrence of many diseases is closely related to the systems,especially cardiovascular,cerebrovascular and thrombotic diseases and coagulation factor deficiency diseases.Now,coagulation function test is mainly divided into conventional coagulation function and thromboelastography test.Generally,the conventional coagulation function test detect a part of the coagulation chain reaction in isolated plasma,and generate one point as a result.While the thrombelastography test can observe the whole process of coagulation and fibrinolysis comprehensively and provide information about the properties and dynamics of coagulation and fibrinolysis.Domestically,the American thromboelastography device is the main method for the whole blood coagulation,but complicated mechanical structure and difficult processing of drape wire are the significant problem.In view of the limitations of these two detection methods,this thesis proposes a thrombelastography coagulation detection method based on magnetic beads reciprocating motion,which is referred to as rotating magnetic beads method.This thesis indicates the detection principle of blood coagulation instrument and whole blood coagulation analyzers in detail domestically and globally,then analyzes the viscoelastic changes of blood in the process of blood coagulation,and proposes a detection method to reflect the whole blood coagulation process and draws the thrombelastogram through the rotating reciprocating motion track of magnetic beads in the detection cup.Beside the detection method,the corresponding mathematical model is constructed,and the force of the magnetic bead in the test cup is further analyzed;then the theoretical formula of the force moment of the magnetic bead is deduced.After that,the system test scheme is designed,and the experimental platform is built.Then the original signal collected is processed,and the result is obtained according to the envelope curve,in which obtains the corresponding parameters of thromboelastography.According to the theoretical formula and experimental results,the influence factors of magnetic beads on the experimental platform are determined.In order to get better experimental parameters to reflect the blood coagulation process,this thesis has done lots of orthogonal experiments and the optimal experimental scheme is obtained with scientifical arrangement and analysis of the factors.Finally,the experimental device of this subject uses the optimal scheme and thromboelastography to carry out the comparative verification experiment,using the experimental material quality control of thromboelastography and anticoagulant pig blood whole blood respectively.Comparing the R value,K value and angle value measured by the rotating magnetic bead method with the TEG equipment,it is preliminarily proved that the rotating magnetic bead method has the ability to detect the blood coagulation process,and can reflect the coagulation viscoelasticity and other related characteristics from the envelope curve.
