Coagulation is a complex dynamic physiological process,which is the result of coordinated hemostasis of plasma and blood cells during the initial stage of wound healing.However,coagulation disorders can lead to serious pathological complications,such as hemophilia or cardiovascular and cerebrovascular diseases.Currently,coagulation testing is an important method of diagnosing and treating hemorrhagic or thrombotic diseases.During surgery,blood coagulation testing is required to diagnose potential bleeding causes and predict the risk of bleeding during surgery.In the postoperative stage,patients should receive anticoagulant treatment to prevent thrombosis and ensure that there is no excessive anticoagulation through blood coagulation testing.During treatment,blood coagulation testing results are also required as a reference standard for drug dosage adjustment plans.With the increasing demand for point-of-care coagulation testing in clinical practice,research on coagulation detection based on microfluidic technology has also developed rapidly in recent years.However,existing point-of-care testing technologies still have problems such as complex operation,high detection cost,and complex processing technology.Therefore,the development of low-cost,easy-to-use,and rapid measurement of blood coagulation point-of-care testing devices is of great significance for the diagnosis and treatment of diseases based on coagulation disorders.In order to solve the above problems,this article uses silverpolydimethylsiloxane(Ag-PDMS)conductive elastic material as electrode material to achieve integrated molding of electrode flow channels,and develops a stable,reliable,and high yield chip.By combining mechanical and electrical testing methods,it overcomes the shortcomings of existing testing technologies such as high cost,cumbersome instruments,and complex operations,and achieves point-of-care testing of blood coagulation.In this paper,firstly,the material properties of silver polydimethylsiloxane(Ag-PDMS)were studied,the fabrication ratio of electrode materials was determined,and the chip size was determined through finite element simulation;Then,the electrical characteristics of the chip were researched through impedance spectrum experiments;After that,experiments were conducted on the chip using glycerol solution to verify the feasibility of converting electrode deformation into impedance changes.Finally,the feasibility of the chip for blood coagulation detection was verified through whole blood experiments.This low-cost,easy-to-use,and rapid measurement point-of-care blood coagulation testing chip not only plays an important role in diagnosing and treating diseases and adjusting drug doses,but also has the potential to develop into high-end medical devices that detect blood coagulation.
