In the artificially cultivated perennial with the increase of cultivation years,root diseases have become more and more serious.At the same time,the abuse of pesticides and chemical fertilizers has led to a continuous decline in quality and efficacy,which has seriously affected the yield and quality of Atractylodes chinensis and the safety of medicine.In order to ensure the continuous use of Chinese medicine resources,relevant national laws and regulations point out that the breeding of fine seedlings is the core part of the standardized production of Chinese herbal medicines,and the mycorrhizal seedling breeding is one of the effective ways to improve the disease resistance,high quality and high yield of medicinal plants in the later stage.This research will address the problems of reduced effective components of artificially cultivated Atractylodes chinensis,frequent occurrence of diseases,and unstable yield.By analyzing the diversity of soil AM fungi in the root zone of the authentic medicinal material Atractylodes chinensis and separating and screening the dominant native AM fungi,they were inoculated into the rhizosphere of Atractylodes chinensis seedlings to explore the growth-promoting effect of the dominant native AM fungi on Atractylodes chinensis seedlings and accumulation of secondary metabolites The establishment of a mycorrhizal seedling method for medicinal plant Atractylodes chinensis.The main findings are as follows:(1)Atractylodes chinensis,a genuine medicinal material of Chengde,Hebei,can form a good symbiosis relationship with AM fungi.The Glomus is the dominant genus of soil arbuscular mycorrhizal fungi in the root zone of Atractylodes chinensis in this area,and the root zone of wild Atractylodes chinensis soil AM fungi resources are more abundant than cultivation.The difference of soil nutrients in the root zone of wild and cultivated Atractylodes chinensis is one of the important reasons for the difference in AM fungal community structure.Among them,the content of soil organic matter and available phosphorus are the main soil environmental factors that cause the difference in AM fungal community structure in the soil of Atractylodes chinensis in authentic areas.Boron is a trace element that promotes the diversity and abundance of AM fungal communities.(2)Dominikia iranicum was isolated from wild rhizosphere AM fungi of Atractylodes chinensis by molecular biological tech.(3)The effect of AM fungi on the seedlings of Atractylodes chinensis.The results showed that the inoculation of AM fungi significantly increased the biomass of Atractylodes chinensis.The dependence of Atractylodes chinensis on mycorrhiza reached 16%and 32%,respectively.Among them,the fresh and dry weight of the whole plant increased by 34.71%and 47.61%respectively between the following year of inoculation and the non-inoculation,both reaching significant levels.At the same time,the root length,root surface area,average root diameter and root volume of the secondyear Atractylodes chinensis inoculated with AM fungus were significantly higher than those without inoculation;inoculation with AM fungus can increase the relative content of atractylodin in the fibrous roots ofAtractylodes chinensis,compared with inoculation 50.82%.(4)The effect of AM fungi on the seedling growth of Atractylodes chinensis rhizome.The results showed that the inoculation of dominant native AM fungi on the infection and growth promotion was more significant,the dependence of Atractylodes chinensis on it was 28%,and the fresh and dry weight of the whole plant increased by 44.91%and respectively 38.46%.At the same time,the inoculation of AM fungi significantly increased the leaf photosynthetic rate,root vitality and seedling index.Compared with CK,inoculation with GM and OAM increased the root activity by 38.62%and 45.25%,respectively.The strong seedling index of Atractylodes chinensis inoculated with AM fungus was better than that without inoculation.Among them,the inoculation between GM and CK did not reach a significant level,but between OAM and CK Reached a significant level.Therefore,the inoculation of dominant indigenous bacteria promotes the physiological indicators of the rhizome of Atractylodes chinensis more significantly.In conclusion,the soil AM fungi resources in the wild Atractylodes chinensis root zone in the genuine area are richer in cultivation.Induced,isolated and cultivated dominant native AM fungi and Atractylodes chinensis mycorrhizal seedlings have the best combined effect.This combination is One of the key factors of locality to improve the traditional Chinese herbal medicine.
