Growth arrest and DNA damage-45 alpha(Gadd45?)is a small molecular protein of the growth arrest and DNA damage family,whose expression is regulated by a variety of molecules at different cellular levels.After activated,Gadd45? is involved in cell cycle arrest,DNA repair and mitochondrial apoptosis pathways through interactions with a variety of cellular proteins,which is associated with cell proliferation,migration and apoptosis.Embryo implantation is essential for successful pregnancy,which is regulated by various hormones,cytokines and other factors during pregnancy,involving cell proliferation,migration and apoptosis.In this study,mouse models of early normal pregnancy,pseudopregnancy,delayed implantation,activated implantation and artificial decidualization were established.The localization and expression of Gadd45? mRNA in the uterus of the above models were examined by frozen sections and in situ hybridization,and the relative expression levels of Gadd45? mRNA in the uterus of early pregnancy was detected by real-time RCR.The results of in situ hybridization showed that Gadd45? mRNA was expressed mainly in the luminal and glandular epithelium of the uterus during the days 1-4 of early pregnancy,and expressed in the decidual region of the uterus during the days 5-8,with a clear change in the location of expression;In the pseudopregnancy,delayed implantation,activated implantation and artificial decidualization models,the expression is consistent with the normal pregnancy model;the results of real-time PCR indicated that Gadd45? mRNA changed regularly with the process of implantation.The experimental results showed that the Gadd45? gene fragment was successfully cloned and the mouse models of normal pregnancy,pseudopregnancy,delayed implantation,activated implantation and artificial decidualization were successfully established.Gadd45? was expressed in the early normal pregnant mouse uterus,showing spatial and temporal specificity,and its relative expression also changed with the process of embryonic implantation,thus its expression changes in the mouse uterus were related to the process of embryonic implantation.
