Background:In recent years,studies have shown that extracellular vesicles of human adipose-derived stem cells(Human adipose-derived stem cell extracellular vesicle,hADSC-EVs)have a broad application prospect in tissue repair and regeneration.As a new type of acellular therapy for tissue regeneration,EVs can largely avoid the risk of transplantation complications and at the same time exert the same therapeutic effect as stem cells.In this study,Waller's degeneration model was used to evaluate the effect of hADSC-EVs on Schwann cells,focusing on the effects of hADSC-EVs on dedifferentiation,proliferation and apoptosis of Schwann cells in the early stage of Waller degeneration.Methods:The experiment is mainly divided into two parts:(1)to identify the characteristics of isolated and cultured adipose stem cells and extracellular vesicles.(2)the rat model of Waller's degeneration in vitro was prepared and treated with different concentrations of(0ug/ml,50ug/ml,and 100ug/ml)hADSC-EVs for different time(12h,24h).After collecting the model tissues,the dedifferentiation of Schwann cells was evaluated by immunofluorescence staining,Western blotting and Real time PCR.After 24 hours of intervention with different concentrations(0ug/ml and 100ug/ml),the proliferation of Schwann cells was detected by immunofluorescence staining and Real time PCR.Apoptosis was detected by Tunel staining and Western Blot,and the expression of cytokines secreted by Schwann cells was detected by Elisa after different concentrations(0ug/ml,50ug/ml,and 100ug/ml)hADSC-EVs were intervened for different time(12h,24h).Results:ADSCs,was successfully isolated and cultured and its extract was identified to meet the standard of extracellular vesicles.HADSC-EVs can promote the dedifferentiation of Schwann cells in the early stage of Waller's degeneration,promote the proliferation of SCs and inhibit its apoptosis.HADSC-EVs also promotes the secretion of a variety of neurotrophic factors by SCs in the early stage of Waller's degeneration.Conclusion:HADSC-EVs promotes the dedifferentiation of Schwann cells during Waller's degeneration,promotes its proliferation and inhibits its apoptosis,and promotes its secretion of a variety of neurotrophic factors.Thereby forming a microenvironment that is conducive to nerve regeneration.
