Experimental Research Of Promoting Survival After Adipose-stem Cell Transplantation And Survival Of Autologous Fat Transplantation | | Posted on:2020-02-06 | Degree:Master | Type:Thesis | | Country:China | Candidate:M A Luo | Full Text:PDF | | GTID:2370330602953468 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Part ⅠExperimental Research of Chitosan Scaffold Loaded with SHH and CM-Dil Labeled Adipose Derived Stem Cells Survival Differentiation after TransplantationObjective(s):The aim of our research is to observe the survival and differentiation of adipose Derived stem cells after transplantation with injectable chitosan scaffold loaded with SHH and adipose stem cells.Methods:Thirty healthy C57BL/6 mice were selected.Random division group A mixture of PBS and ADSCs,group B mixture of SHH and ADSCs,group C chitosan scaffold loaded with ADSCs,group D chitosan scaffold loaded with SHH and ADSCs.After 1,2,4,8 weeks analysis involved general observation,HE staining,immunohistochemical staining of the survival and differentiation of transplanted cells.Results:Eight weeks after operation,chitosan scaffold were completely degraded.After 8 weeks,analysis involved general observation,HE staining and immunofluorescence staining showed that Chitosan scaffold loaded with SHH and ADSCs could effectively increase the survival rate of adipose stem cells,increase angiogenesis.All the results were better than that of A,B,C group.Conclusion(s):Chitosan scaffold loaded with SHH and ADSCs can effectively promote the survival of stem cells and spontaneously differentiate into mature adipocytes and increase the number of vascular tissues.May serve as a promising strategy for improving the survival of ADSCs graft survival rate.Part ⅡExperimental Research of hydrogen sulfide Enhancing the Survival Rate of Fat TransplantationObjective(s):To explore the concentration of hydrogen sulfide in blood could affect the survival rate of autologous fat transplantation.Methods:Eighteen BALB/c mice were divided into three groups:Blank group(fat granules only),H2S group(NaHS were injected in abdominal cavity everyday after transplanting the fat granules),SHH group(mixture of SHH and fat granules).Six mice in each group.Mixture was injected into the subcutaneous tissue in the back of mice respectively.The mice were killed in the 1st,2nd,4th week after the operation.Transplanted tissues come from operations were analyzed by means of morphological observation,apoptosis experiment,flow cytometry,HE-staining and immunohistochemical technology.Results:According to the apoptosis experiment and flow cytometry,the apoptosis rate of each group is:Blank group>H2S group>SHH group in the first week;Blank group>H2S group and SHH group in the second week.According to the cell counting,the number of H2S group and SHH group were greater than Blank group.According to the observation,HE staining and immunohistochemical staining,the H2S could improve the survival rate of transplanted tissue,reduce the cell apoptosis rate and promote the proliferation of new blood vesselConclusion(s):Increasing the concentration of blood H2S can reduce the necrosis rate of transplanted tissue and promote the formation of blood vessel effectively. | | Keywords/Search Tags: | syringeability, chitosan, SHH, adipose stem cells, stem cell transplantation, Hydrogen sulfide, Fat Transplantation, Mice, Apoptosis, Angiogenesis | PDF Full Text Request | Related items |
