Due to the absence of neurovascular distribution,of cartilage lesions,it is extremely difficult,once damaged,for cartilage to get regeneration.For the regeneration of damaged cartilage,cartilage tissue engineering and cartilage regeneration medicine provides a new treatment option,and cast a hope to the patients.Seeding cells are the hot spots of cartilage tissue engineering,and attract a large number of scholars to carry out relevant research.Due to limited quantity of autologous chondrocytes,which are belonged to weakly proliferating quiescent cells.Besides,chondrocytes tend to get dedifferentiation to loss their capacity to form normal cartilage tissue.Because of strong proliferating capacity,multi-directional differentiation potential,stem cells inspired lots of research and exploration.Bone marrowderived mesenchymal stem cells(BMSCs)have been widely and deeply studied,but a large number of studies have shown that the cartilage tissue,formed by the chondrogenic differentiated bone marrow mesenchymal stem cells,tends to lose the stable cartilage phenotype,gradually get fibrosis,vascularization and even ossification.As research progresses,one subtype cells with pluripotent properties are found in cartilage tissue,and these cells may become the ideal seeding cells for tissue-engineered cartilage.In this study,newborn pig auricle cartilage derived stem cells have been chosen as the research object,preferred by histological means to explore the histological characteristics and localization of the auricle cartilage tissue and perichondrogenium derived stem cells;followed by the method of differential adhesion,we harvested the stem cells from auricle cartilage tissue and perichondrogenium.We imployed flow cytometry to detect stem cells surface markers,clone formation assay to test their proliferative capacity;in vitro,we test their osteogenic,adipogenic and chondrogenic differentiation to explore their multidifferentiated ability;last,we selected BMSCs as control group,we seed these two subtype stem cells onto biological scaffolds to explore their ectopic cartilage formation capacity.
