Under normal circumstances,the generation and elimination of free radicals in the body is in a state of balance.However,in the exhaustive movement,a large number of free radicals are generated in the body,and the equilibrium is broken,causing free radical accumulation.The accumulated free radicals attack the body's biomolecules,cells,glands and tissues,causing irreversible injury,damaging the body and causing diseases and aging.In order to maintain the balance between the production and elimination of free radicals in exhaustive exercise,the body needs to ingest some essential antioxidants to enhance the activity of antioxidant enzymes and improve the body's ability to scavenge free radicals.Plant polyphenols,as green,natural,non-toxic and highly effective antioxidants,have become the first choice for this study.Dendrobium candidum is a precious Chinese herbal medicine that has the functions of enhancing immunity,nourishing yin,replenishing spleen and stomach,protecting liver,protecting gallbladder,strengthening bones,lowering blood sugar,and inhibiting tumors.Dendrobium candidum contains a variety of active ingredients,of which polyphenols have good antioxidant effects in enhancing the body's antioxidant enzyme activity,and improving the ability to scavenge free radicals.Therefore,the body should be supplemented with dendrobium candidum polyphenols before training to effectively prolong the exercise time and improve the exercise capacity.It can also be supplemented with dendrobium candidum polyphenol after exercise training to timely remove the free radicals generated by high-intensity exhaustive training in the body to prevent oxidative damage caused by free radicals.OBJECTIVE: This experiment used dendrobium candidum polyphenols.Each experimental mouse was given 200 mg/kg dendrobium candidum polyphenol extract before exercise training.After four weeks of exercise training,the serum was measured for levels of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px)and lipid peroxide dialdehyde(MDA)content in each group of mice.In order to reflect the anti-oxidation effect of dendrobium candidum polyphenol extract on exhausted exercised mice,healthy and effective natural antioxidant drinks were chosen for health guidance in exercise training,post-exercise rehabilitation,releasing exercise fatigue and improving exercise capacity.METHODS: In this study,dendrobium candidum was collected from Jinan Hong Kong-based flower “Meiyuan Boutique No.1”,where it was frozen to protect the integrity of the molecular structure of each substance.The polyphenols of dendrobium candidum were extracted with ultrasonic assisted and heated bath,using ethanol as the extraction solvent.The content of polyphenols in the extract was determined by using gallic acid standard to be 103.5 mg/g,and the content of polyphenolic flavonoids in the extract was determined to be 24.8 mg/g.90 healthy male Kunming mice were purchased from the Animal Experimental Center of Shandong University of Traditional Chinese Medicine.According to the Chinese Pharmacopoeia,the daily dose of dendrobium candidum was 6-12 g.According to the 60 kg and the minimum dosage(6g)of an adult body,the dosage of dendrobium candidum should be 100.00 mg/kg · d and the design conditions of pre-experimental dose.50,100,200,300,500 mg / kg of groping,the experimental mice were determined to be given a dose of 200 mg / kg.Referring to the reviewed literature,the first week of mice swimming exercise training volume was 30 min,the second week was 60 min,the third week was 90 min,the fourth week was 120 min and the last swimming exercise was to force exhaust.The mice were killed by eyeball retrieval and serum was separated.The elisa kit was provided by the Enzyme-free Reagent Company,and according to the strict requirements of the kit,the operation was carried out carefully,the absorbance was measured by the enzyme analyzer,the standard curve was drawn,and the enzyme content was calculated by the regression equation.RESULTS: SOD enzyme: The serum SOD values in the PL group were significantly higher than those in the NC group,with a significant difference(P<0.001).The serum SOD content in the PL group was higher than that in the PC group,and there was a significant difference(P<0.01).The serum SOD content in the N-S group was significantly higher than that in the NC group,and there was a significant difference(P<0.01).The serum SOD content in the P-S group was higher than that in the PC group,and there was a significant difference(P<0.01).CAT enzyme: The serum CAT content in the PC group was significantly higher than that in the NC group,and there was a significant difference(P<0.001).The serum CAT content in the PL group was significantly higher than that in the NC group,and there was a significant difference(P<0.001).There was no significant difference in serum CAT content between the PC group and the PL group(P>0.05).The serum CAT content of the mice in the P-S group was significantly higher than that in the N-S group,with a very significant difference(P<0.001).The CAT content in the N-S group was significantly higher than that in the NC group,which was very significant(P<0.001).The CAT content in the P-S group was significantly higher than that in the PC group(P<0.01),which was significantly different.GSH-Px enzyme: The serum GSH-Px enzyme content in the PC group was higher than that in the NC group(P<0.001),which was very significant.The serum GSH-Px enzyme content in the PL group was significantly higher than that in the NC group(P<0.01),which was significantly different.There was no significant difference in the serum GSH-Px enzyme content between the PC group and the PL group(P>0.05).The serum GSH-Px enzyme content in the N-S group was significantly higher than that in the NC group(P<0.001),which was very significant.The serum GSH-Px enzyme content in the P-S group was significantly higher than that in the PC group(P<0.01),which was significantly different.The serum GSH-Px enzyme content in the Vc exhausted exercise group was higher than that in the PC group after 24 hours(P<0.05),which was significantly different.MDA content: Compared with NC,the serum malondialdehyde content in the N-S group mice was higher than that in the NC group,and the difference was significant(P<0.01).The serum malondialdehyde content in the PC group was higher than that in the NC group,and the difference was significant(P<0.05).The serum malondialdehyde content in the PL group was higher than that in the NC group,and there was a significant difference(P<0.001).The serum malondialdehyde content in the N-S group was higher than that in the NC group,and there was a significant difference(P<0.01).Conclusion: Dendrobium candidum polyphenols significantly increased the levels of superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px)in the mice serum,enhancing their exercise capacity,and extending their swimming time.The 4-week long exercise training and the final exhaustive exercise did not reduce the level of antioxidant enzymes,but increased the antioxidant enzyme content.Therefore,the experimental results show that supplementation of dendrobium candidum polyphenols and swimming training have both enhanced the antioxidant capacity of mice.
