stability.Injury of the multifidus muscles of the waist leads to instability of the lumbar spine,weakening of the waist function and even causing low back pain,which seriously affects people’s daily work.Therefore,the repair of lumbar multifidus muscle injury is worthy of attention.Mesenchyal stem cells(MSCs)in the microenvironment of the lumbar multifidus muscle can promote the repair of the multifidus muscle after the injury by the paracrine effect of secreted cytokines spreading to adjacent cells Hepatocyte growth factor(HGF)as a paracrine factor of MSCs plays a significant role in the change of skeletal muscle injury microenvironment.Therefore,this study observes the changes and rules of HGF-related factors secreted by MSCs by electroacupuncture"Commission",and provides some experimental basis for the repair of lumbar multifidus muscle injury.PurposeTo study the effect and regularity of electroacupuncture "Commission" on the secretion of HGF and related factors in the distally injured multifidus muscle to promote the repair of lumbar multifidus muscle injury.Method.54 male SD rats were randomly divided into blank group,model group and electroacupuncture group,18 rats each.The model group and the electroacupuncture group were injected with four points on both sides of the L4 and L5 levels of the bilateral spine,and 0.5%bupivacaine solution was used.It was fixed 24 hours after the model was built.The electro-acupuncture group intervened on both sides of the "Via Zhong" acupuncture point.The Korean-style electro-acupuncture instrument HANS-200A was used.The sparse and dense wave of 2/100 Hz,the current intensity was 1 mA,once a day,once every 20 minutes,To 1 day before taking the material.The blank group was fed normally without treatment.All the three groups were synchronously selected after 1 day,3 days and 7 days of treatment.The expression of HGF,HGFA and c-Met in the multifidus muscle of the waist was observed by enzyme-linked immunosorbent assay(ELISA),and HGF and c were measured by immunohistochemistry-Myc positive expression,Western Blot detection c-Met expression changes.Result1 HE stained muscle fiber morphology resultsBlank group:Muscle fibers are slender and multi-nucleated,unifonn in thickness and tightly distributed in the longitudinal direction;nuclei are oblong and close to the sarcolemma.After 1 day of intervention,the muscle fibers of the model group were necrotic and deformed,and the gap between the muscle fibers was significantly widened,with a large number of inflammatory cells distributed in the gap;the muscle fibers in the electroacupuncture group were widened and the gap was shorter than the model group,and the number of infiltrating inflammatory cells was reduced.After 3 days of intervention,the shape of the muscle fibers in the model group was still irregular,and some of the arrangement began to be compact,but there were still fragments of muscle fibers and a large number of inflammatory cells;the muscle fibers in the electroacupuncture group were more uniform and regular in arrangement,and the gap was still inflammatory Cell infiltration.After 7 days of intervention,new muscle fibers were seen in the model group,and the gap between the fibers was significantly shorter than that in the model group of 1 day and 3 days of intervention,and some inflammatory cells were expression changes of HGF,HGFA and c-Met①HGF:Compared with the blank group,there was no statistically significant difference in the expression of HGF in the model group after treatment compared with group,the expression level of HGF in the electroacupuncture group was higher than that in the model group on one day(p<0.01);the expression levels of HGF in the electroacupuncture group on the 3 and 7 was higher than that in the model group(p<0.01).③ c-Met:Compared with the blank group,the expression of c-Met of the multifidus muscle in the model group was lower than that of the blank group(p<0.05);the expression of c-Met of the multifidus muscle in the model group was less than that of the blank group.In comparison,the difference was not statistically significant(p>0.05);the expression level of c-Met in lumbar multifidus muscle in the model group was lower than that in the blank group(p<0.05).In the electroacupuncture group,the expression level of c-Met in the lumbar multifidus muscle was low in the electroacupuncture group and the blank group(p<0.01).The expression level of c-Met in the multifidus muscle of the waist group was lower in the needle group than in the blank group(p<0.05).Compared with the model group,there was no statistically significant difference in the expression of c-Met in the lumbar multifidus muscle between the electroacupuncture group on day 1 and day 7(p>0.05);Model group(P<0.01)3.Immunohistochemical determination of HGF and c-Myc positive expression①HGF:Comparing the model group with the blank group,the HGF positive expression that of the blank group(p<0.01).Compared with the model group,the positive expression values of c-Myc in the lumbar multifidus muscle in the electroacupuncture group were higher than that in the 1st and 3rd days,but the difference was not statistically significant(p>0.05);the 7th lumbar multifidus muscle c in the electroacupuncture group-Myc positive expression was higher than the model group(P<0.01).4.WB measured c-Met expressionCompared with the blank group,the c-Met expression of the lumbar multifidus muscle in the model group was lower than that in the blank group,and the
