ObjectionIn recent years,with the development of intestinal flora sequencing technology,the relationship between intestinal flora and disease has been paid more and more attention.As one of the chronic complications of diabetes,diabetic nephropathy brings a great burden to both individuals and society.The treatment effect of traditional Chinese medicine on diabetic nephropathy is definite and widely recognized and affirmed.Studies have shown that there are certain differences in the composition and types of intestinal microflora in patients with diabetic nephropathy and normal healthy people.After preliminary investigation and research,it is shown that the diabetic nephropathy damp-heat syndrome is the most common type of diabetic nephropathy in the early and middle stages,but the reasons for the damp-heat syndrome are diverse.This study aims to explore the possible causes of the formation of diabetic nephropathy damp heat syndrome by exploring the differences in the composition and distribution of intestinal microflora in the diabetic nephropathy damp heat syndrome and non-damp heat syndrome.MethodsA total of 24 inpatients with diabetic nephropathy in the Department ofNephrology of Dongzhimen Hospital were collected,including 12 cases of damp heat syndrome and 12 cases of non-humid fever syndrome.The patient's condition data and intestinal flora specimens were collected respectively,through bacterial DNA extraction,PCR amplification,and Miseq library construction And Miseq analyzed the intestinal microflora samples of the two groups after the computer sequencing process.SPSS 26 software was used for data statistics.Among them,general data such as gender and age were tested by T test.Intestinal flora analysis was based on OTU basic analysis,Alpha diversity analysis,species composition analysis,and Beta diversity analysis.-Whitney rank sum test(MW test)or Kruskal-Wallis rank sum test,hypothesis test with P<0.05 is considered statistically significant.ResultsThis sequencing generated a total of 981 OTUs,including 699 damp heat syndrome groups and 838 non-humid fever syndrome groups.The results of OTU basic analysis,Alpha diversity analysis,species composition analysis,Beta diversity analysis,and statistical tests on the two groups of samples are as follows:1.There is no significant difference in the number of OTUs between the two groups;2.Group Chao 1 index,PD-Whole-tree index,Shannon index,Simpson index and other analysis,there is no significant difference in Alpha diversity between the two groups;4.Analysis of the intestinal flora species composition of the two groups,combined with Kruskal-Wallis rank After the test,it was found that there were significant differences in the composition of the intestinal flora between the two groups,specifically as follows:Aminococcaceae(P=0.003),koalabacteria(P=0.014),active gastrinoma(P=0.01),Eagle's genus(P=0.042),galactanoic acid bacteria(P=0.024),Sellimonas bacteria(P=0.037),Lactonifactor bacteria(P=0.014),Lachnoclostridium Bacteria(P=0.033)and other bacteria are increasing;Proteobacteria(P=0.014),Campylobacteria(P=0.014),Prevotellaceae(P=0.003),Lactobacillus(P=0.004),Pasteurellaceae(P=0.042),Micrococcaceae(P=0.037),Campylobacteriaceae(P=0.014),Prevotella-9(P=0.018),Fecal bacteria(P=0.021),Lactobacillus(P=0.003),Sartre(P=0.04),Haemophilus(P=0.042),Lactococcus lactis(P=0.046),Enterobacter(P=0.033),Leuconostoc(P=0.033),Rossella(P=0.037),Campylobacter(P=0.032),Senegalimassilia genus(P=0.015),Pasteurellales genus(P=0.042)and other genus decreased;5.After Spearman correlation analysis,it can be seen that the bacteria caused by the difference of dampness and heat are Proteobacteria,Campylobacter,Prevotidae,Aminococcaceae,Campylobacteraceae,Prevotella 9.Fecal bacillus,Lachnoclostridium fungus,Koala bacillus,active rumenococcus,rumen fungus-UCG-003,Senegalimassilia fungus Among them,Prevotellaceae belong to strong correlation(|?|=0.619),and the rest of the flora belong to moderate correlation.Conclusions1.Through sequencing analysis of intestinal micro flora in patients with diabetic nephropathy damp heat syndrome and non-humid fever syndrome,combined with Kruskal-Wallis rank sum test,we can see that there are significant differences in the composition of certain flora in the damp heat group and the non-humid heat group2.Combined with the Spearman correlation coefficient,it can be seen that among the different bacterial groups,the bacteria related to the heat and humidity are Proteobacteria,Campylobacter,Prevotidae,Aminococcus,Campylobacteria,Prevot Bacteria-9,Fecal bacteria,Lachnoclostridium bacteria,Koala bacteria,active rumenococcus,rumen bacteria-UCG-003 and Senegalimassilia genus,the content of these flora may be changed due to the evil of damp heat Thereby producing a group of symptoms of damp heat.
