| Objective:1.To discuss the objectivity of "observing liver diseases,knowing liver diseases can transmit to spleen,and strengthening the spleen first" based on the rat models of liver depression,and liver depression combined with chronic liver injury.2.To discuss the biological mechanism of "observing liver diseases,knowing liver diseases can transmit to spleen,and strengthening the spleen first" from the perspectives of hepatocyte apoptosis and HPA axis based on the rat model of liver depression combined with chronic liver injury.Methods:1.Pharmacodynamic Study on "observing liver diseases,knowing liver diseases can transmit to spleen,and strengthening the spleen first":Based on pharmacodynamic research methods,the rat models of liver depression and liver depression superimposed with chronic liver injury are duplicated respectively.The treatment method of "strengthening the spleen first" is duplicated through different groups and successive dosing methods(soothing liver,invigorating spleen,soothing liver and invigorating spleen,soothing liver and invigorating spleen first,soothing liver and invigorating spleen later,soothing liver and invigorating spleen first,soothing liver and invigorating spleen later).Western blot,RT-PCR and other molecular biological detection methods were used to explore the biological mechanism of "observing liver diseases,knowing liver diseases can transmit to spleen,and strengthening the spleen first" from the perspective of hepatocyte apoptosis and HPA axis.Results:1.Pharmacodynamic Study on "observing liver diseases,knowing liver diseases can transmit to spleen,and strengthening the spleen first"First of all,there are different degrees of abnormal changes in the general situation,body mass change trend,serum liver function biochemistry,liver histopathology,gastrointestinal mucosal barrier and gastrointestinal hormone indexes of rats with liver depression and rats with liver depression combined with chronic liver injury."Observing liver diseases,knowing liver diseases can transmit to spleen" objectively exists in the above two models.All intervention methods(soothing the liver,invigorating the spleen,soothing the liver and invigorating the spleen,first soothing the liver,then soothing the liver and invigorating the spleen,first invigorating the spleen,then soothing the liver and invigorating the spleen)have different degrees of intervention effects on the above indicators.The intervention effect of"strengthening the spleen first" on the above indicators is better than that of are differences in the detection results of liver function biochemical index ALT and gastrointestinal hormone index CCK between liver depression rats and liver depression plus chronic liver injury "observing liver diseases,knowing liver diseases can transmit to spleen,and strengthening the spleen first".2.Study on the biological mechanism of "knowing the disease by seeing the liver,knowing the liver disease will be transmitted into the spleen,strengthening the spleen first" from the perspective of hepatocyte apoptosis and HPA axisTUNEL fluorescence scanning results of rats with liver depression combined with chronic liver injury showed that apoptotic cells increased significantly under different visual fields.AI increased significantly;The expression of Bax gene and Bax/Bcl-2 were significantly higher than those in normal group,while the expression of Bcl-2 gene was down-regulated,but the difference was not significant.The relative expression of Bax and Bax/Bcl-2 protein increased significantly,while the expression of Bcl-2 protein decreased but not significantly.The contents of hypothalamic CRH and serum ACTH and CORT increased significantly.Compared with the model group,each dosing group has different degrees of positive regulation on the above indexes,and Chaishao Liujunzi Decoction group C,which replicates "strengthening the spleen first",has the best comprehensive regulation on each index.Conclusions:1.From the pharmacodynamic point of view,"knowing the disease by seeing the liver,knowing the liver disease will be transmitted into the spleen" and " knowing the liver disease will be transmitted into the spleen,strengthening the spleen first" have scientific objectivity.2."Strengthening the spleen first" has a significant regulatory effect on hepatocyte apoptosis.Through the direct regulation of hepatocyte apoptosis,the expression of pro-apoptotic protein/gene Bax can be down-regulated and the expression of anti-apoptotic protein/gene Bcl-2 can be up-regulated,which may be one of the biological mechanisms of"knowing the disease by seeing the liver,knowing the liver disease will be transmitted into the spleen,strengthening the spleen first".3.After being absorbed by the digestive system,the medicine of "strengthening the spleen" can reduce the secretion of hypothalamus CRH through negative feedback mechanism of HPA axis while preventing and treating "spleen disease" by reducing the content of CORT in the peripheral system,thus reducing the secretion of ACTH,further reducing the secretion of CORT and inhibiting the transcription of relevant inflammatory genes,thus achieving the effect of effectively treating liver diseases.It may be one of the biological mechanisms of"knowing the disease by seeing the liver,knowing the liver disease will be transmitted into the spleen,strengthening the spleen first". |
