Purpose:To investigate the effect and mechanism of Xiangsha Liujunzi decoction on liver cell pyrolysis in rats with hyperlipidemia.Material and method:75 SD rats were randomly divided into normal group,high fat group,spleen deficiency and high fat group,Xiangsha Liujunzi Decoction high dose treatment group and Xiangsha Liujunzi Decoction middle dose treatment group after two weeks of adaptive feeding with common feed.The rats of spleen deficiency and high fat group,Xiangsha Liujunzi Decoction high dose treatment group and Xiangsha Liujunzi Decoction medium dose treatment group were used to model spleen deficiency for 15 days.The rats of high fat group,spleen deficiency and high fat group,Xiangsha Liujunzi Decoction high and medium dose group were fed with high fat feed for 14 weeks.The rats of normal group were fed with normal diet.At the 10 th week,Xiangsha Liujunzi Decoction The middle dose group and the high-dose group began to infuse the stomach with traditional Chinese medicine,and the middle dose treatment group of Xiangsha Liujunzi Decoction was 5.67g/kg A kind of d)The dosage of Xiangsha Liujunzi Decoction was 11.34 /(kg)A kind of d)To the stomach.Normal group,high-fat group and spleen deficiency high-fat group were administrated with corresponding volume of normal saline for 4 weeks.Four weeks after administration,total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C)in serum of rats in each group were detected by automatic biochemical analyzer,and the excretion rate of D-xylose in urine of rats in each group was checked;the liver pathomorphology of rats in each group was observed by HE staining and oil red O staining The changes of IL-1β,IL-18,gsdmd,NLRP3,caspase-1 m RNA,gsdmd,NLRP3 and caspase-1 m RNA,NLRP3 and caspase-1 m RNA,NLRP3 and caspase-1 m RNA,NLRP3 and caspase-1 protein,and ELISA protein in liver were detected.Results:1.Effect of Xiangsha Liujunzi Decoction on blood lipid level in rats with spleen deficiency and hyperlipidemia.Compared with the normal control group,the serum TC,TG,LDL-C in the high-fat group and spleen deficiency high-fat group were significantly higher,and HDL-C was significantly lower(P < 0.01).Compared with the spleen deficiency high-fat group,the serum TC,TG,LDL-C in the high-dose treatment group and the middle dose treatment group were significantly lower(P < 0.05 or P < 0.05)HDL-C increased significantly(P < 0.01).2.The effect of Xiangsha Liujunzi Decoction on the pathomorphological changes and lipid deposition of liver in rats with spleen deficiency and hyperlipidemia.HE staining results showed that: compared with the normal control group,the hepatocytes in the high fat group and the spleen deficiency and high fat group were loose,swollen and round,and there were more round vacuoles under the microscope;compared with the liver cells in the spleen deficiency and high fat group,the hepatocytes in the high dose treatment group and the middle dose treatment group were closely arranged,and the degree of swelling was reduced The number of round vacuoles decreased significantly.The results of oil red O staining showed that: compared with the normal control group,there were more red lipid droplets in the cytoplasm of the liver cells of the high fat group and the spleen deficiency and high fat group;compared with the liver cells of the spleen deficiency and high fat group,the red lipid droplets in the liver cells of the middle dose treatment group and the high dose treatment group were significantly reduced.3.Effect of Xiangsha Liujunzi Decoction on the expression of apoptosis related proteins in liver and serum of rats with spleen deficiency and hyperlipidemia.The results of Western blot showed that the expression of NLRP3,gsdmd and caspase-1in the liver of the rats in the high fat group and the spleen deficiency high fat group increased to different degrees compared with the normal control group(P < 0.01 or P < 0.05);compared with the spleen deficiency high fat group,the gsdmd protein in the liver cells in the high dose group and the middle dose group of Xiangsha Liujunzi Decoction decreased to different degrees(P < 0.05 Or P < 0.01).NLRP3 and caspase-1 protein in Xiangsha Liujunzi Decoction high-dose treatment group decreased in varying degrees(P < 0.05 or P < 0.01).The expression of NLRP3 and caspase-1 protein in Xiangsha Liujunzi Decoction quantitative treatment group was not statistically significant,but there was a significant downward trend.The results of ELISA showed that: compared with the normal control group,the expression of IL-18 and IL-1β protein in the liver and serum of the rats in the high fat group and the spleen deficiency high fat group increased to different degrees(P < 0.01);compared with the spleen deficiency high fat group,the expression of IL-18 protein in the liver and serum of the rats in the high dose and the middle dose treatment groups of Xiangsha Liujunzi Decoction decreased to different degrees(P < 0.05 or P < 0.01).The expression of IL-1β protein in the liver and serum of rats in the high-dose treatment group of Xiangsha Liujunzi Decoction was also decreased(P < 0.05 or P < 0.01),and the expression of IL-1β protein in the liver of rats in the middle dose treatment group of Xiangsha Liujunzi Decoction was decreased(P < 0.05).Although the expression of IL-1β protein in the serum was not statistically significant,it had a significant downward trend.4.Effect of Xiangsha Liujunzi Decoction on m RNA of apoptosis related protein in liver of rats with spleen deficiency and hyperlipidemia.The results of Realtime RT-PCR showed that the m RNA of NLRP3,GSDMD,Caspase-1,IL-18 and IL-1β in the liver of rats in the high fat group and the high fat group with spleen deficiency increased in different degrees(P < 0.01).Compared with the high-fat group and spleen deficiency high-fat group,the m RNA of NLRP3,gsdmd,caspase-1,IL-1βprotein in the liver of the rats in the middle dose and high-dose treatment groups of Xiangsha Liujunzi Decoction decreased in varying degrees(P < 0.05)There was no statistical significance in the expression of IL-18 m RNA in the liver of rats in the middle dose treatment group of Xiangsha Liujunzi Decoction,but there was a significant downward trend.Conclusion:1.Xiangsha Liujunzi Decoction can reduce the blood lipid level and improve the liver lipid deposition in rats with spleen deficiency and hyperlipidemia.2.Xiangsha Liujunzi Decoction may improve the condition of liver lipid deposition by changing the genes of apoptosis related protein and apoptosis related protein in the liver of spleen deficiency hyperlipidemia rats.
