Purpose: Based on the screening of medication patterns,this study aims to combine clinical and animal experiments to study the effect of inflammation on the obstruction of lipid transfer in spleen deficiency ointment,as well as the effect of Xiangsha Liujunzi decoction on liver NF-κ B/NLRP3 inflammasome pathway,thereby elucidating the mechanism of Xiangsha Liujunzi decoction in regulating dyslipidemia.Materials and methods:1.By excavating the ingredients of Xiangsha Liujunzi decoction: Ginseng,Atractylodes,Poria cocos,Boiled Licorice,Crow-dipper,Tangerine peel,Aucklandiae Radix,and Amomi Fructus,the medicinal properties,medicinal taste,and drugs are summarized and statistically sorted out,and the pharmacological effects of Xiangsha Liujunzi decoction are excavated.2.According to the inclusion criteria,diagnostic criteria,and exclusion criteria,people with spleen qi deficiency and dyslipidemia and healthy volunteers were recruited,and informed consent forms were signed to analyze the general data such as age,gender,BMI,and TCM symptom scores of the included cases.After 4 weeks of clinical intervention in Xiangsha Liujunzi decoction in people with spleen qi deficiency and dyslipidemia,serum TC,TG,LDL-C,HDL-C levels were detected by automatic biochemical analyzer,and serum VCAM-1,ICAM-1,E-selectin,IL-8,IP-10 and CCL2 contents were detected by ELISA.This study illustrates the effect of Xiangsha Liujunzi decoction on adhesion factors and chemokines in patients with spleen qi deficiency and dyslipidemia.3.SPF SD rats were randomly divided into blank control group,high-fat group,spleen deficiency and high-fat group and Xiangsha Liujunzi decoction group,using the model of spleen deficiency caused by uncontrolled diet and exhaustive swimming,after successful modeling,the blank control group was fed with basic feed,and the rest of the groups were fed with high-fat feed for a total of 14 weeks,and administered by gavage on the 10 th week.The serum levels of TC,TG,LDL-C and HDL-C were detected by automatic biochemical analyzer,and the serum VCAM-1,ICAM-1,E-selectin,IL-8,IP-10,CCL2,IL-6,TNF-α,IL-18,IL-1β and the key factors of liver inflammation IL-6,TNF-α,IL-18 and IL-1β were detected by ELISA.In order to explore the effect of Xiangsha Liujunzi decoction on the inflammatory pathway of rat spleen deficiency and hyperlipidemia,the m RNA levels of IL-6,TNF-α and IL-18 genes were detected by q RT-PCR and Western blot.Results:1.Most of the deficiency tonic drugs in Xiangsha Liujunzi decoction are qi-tonifying drugs,accounting for 37% of the total frequency,followed by qi-regulating drugs,accounting for 25% of the total frequency,and the rest are damp-dissolving drugs,warming cold and phlegm drugs and water-reducing swelling drugs.Xiangsha Liujunzi decoction is mainly warm drugs,accounting for 75% of the total drugs,followed by flat drugs;Among the five flavors,sweet,bitter and pungent drugs were the most,followed by light taste.Among the meridians,the most drugs are returned to the spleen meridian,followed by the lung meridian and the stomach meridian.2.Compared with healthy people,the serum levels of VCAM-1,E-selectin,IP-10,ICAM-1,CCL2 and IL-8 in people with spleen qi deficiency and dyslipidemia were significantly increased before treatment(P<0.01),among which TG showed a strong positive correlation with ICAM-1,and the sensitivity of six chemokines and adhesion factors such as VCAM-1 exceeded 79.4%,the specificity exceeded 89.3%,and the area under the AUC curve was greater than 0.883,which had good diagnostic value for spleen qi deficiency and dyslipidemia.After the intervention of Xiangsha Liujunzi decoction,the blood lipid level of the people with spleen qi deficiency and dyslipidemia was improved,and the serum levels of six chemokines and adhesion factors such as VCAM-1 were significantly decreased(P<0.01).3.Compared with the blank control group,the serum levels of TC,TG,LDL-C in the high-fat group and the high-fat group with spleen deficiency were significantly increased(P<0.01),and the levels of HDL-C were significantly decreased(P<0.01),and the lipid droplet deposition and fat vacuoles in liver tissue were obvious,and the lesions in the high-fat group with spleen deficiency were more obvious,and the serum IL-1β,IL-6,IL-8,IL-18,ICAM-1,VCAM-1,TNF-α,IP-10,CCL2,The contents of IL-1β,IL-6,TNF-α and IL-18 in E-selectin and liver were significantly increased(P<0.01),the m RNA expression of IL-6,TNF-αand IL-18 genes was significantly increased(P<0.05 or P<0.01),and the expression levels of NF-κ B,NLRP3,Caspase-1 and IL-1β genes and proteins were significantly increased(P<0.01),among which the increase trend was more obvious in the spleen deficiency and high-fat group.After the intervention of Xiangsha Liujunzi decoction,the blood lipid disorder was improved,the spleen deficiency state was alleviated,the pathological changes in the liver were restored,the serum IL-1β,IL-6,IL-8,IL-18,ICAM-1,VCAM-1,TNF-α,IP-10,CCL2,E-selectin and IL-1β,IL-6,TNF-αand IL-18 in the liver were significantly reduced(P<0.05 or P<0.01),and the m RNA expression of IL-6,TNF-α and IL-18 genes in the liver was significantly decreased(P<0.01),the expression levels of NF-κB,NLRP3,Caspase-1 and IL-1β genes and proteins in the liver were significantly decreased(P<0.01).Conclusion:1.Research on medication patterns has found that Xiangsha Liujunzi decoction has scientific significance in preventing and treating spleen deficiency and dyslipidemia.2.Xiangsha Liujunzi decoction can effectively improve the blood lipid levels of people with spleen deficiency and dyslipidemia,and its regulatory mechanism may be related to the expression of six chemokines such as VCAM-1 and adhesion factors.3.Xiangsha Liujunzi decoction can effectively improve the blood lipid levels and correct the spleen deficiency state in rats with abnormal blood lipids due to spleen deficiency.Its regulatory mechanism may be related to NF-κ B/NLRP3 inflammasome pathway mediates the expression of VCAM-1 and other inflammatory related factors. |
