Purpose : The Traditional Chinese medicine jieguxujinwan has obvious effects in clinical treatment of fractures,but lacks related research mechanism of modern medicine.In this experiment,we examined the activation status of Wnt/?-catenin signaling pathway in the bone tissue of fractured SD rats,and learned the regulation effect of jieguxujinwan on the signaling pathway during the treatment.;by detecting the level of serum inflammatory factors in fractured SD rats,the control effect ofjieguxujinwan on inflammation was explored;the possible mechanism of action was explored to provide a basis for the follow-up study of the preparation.Materlal and method: Healthy adult male rats were selected and randomly divided into4 groups,10 in each group.There were no significant differences in body weight and age between groups.These groups are: blank group,model group,Chinese medicine group,and control group.In addition to the blank group rats,all rats were anesthetized and all rat femur fracture models were made using fracture scaffold replication.After the model was successfully replicated,both the blank group and the model group were treated with normal saline 10 m L/(kg.d),and once a day.In the control treatment group and the Chinese medicine group,after the model was manufactured,the Chinese patent medicine and the xujinjieguwan10 m L/(Kg.d)liquid were respectively administered.All rats were intragastrically administered once a day for a total of 30 days.After the treatment,the rats were anesthetized with a 5% urethane solution.Blood was taken from the abdominal aorta after laparotomy and serum was taken at low temperature.The levels of serum TNF-? and IL-6 were detected by enzyme-linked immunosorbent assay.The femoral fracture tissue of the rat was taken,and a part of physiological saline was infiltrated,and then stored in an ultra-low temperature refrigerator at-80?.The expression of Wnt/?-catenin key protein in bone tissue was determined by Western Blot.Results:1.The jieguxujinwan significantly improved the general activity status of the fractured rats.The increase of body weight in rats was significantly different from that in other groups(P<0.05).2.Compared with normal rats,the levels of serum TNF-? and IL-6 in the model group were significantly increased(P<0.05),and the inflammatory reaction was obvious.After treatment,the inflammatory response of all rats was alleviated,and the levels of TNF-? and IL-6 were decreased(P<0.05).3.Compared with normal rats,the contents of Wnt3 a and ?-catenin protein in bone tissue of fractured rats were significantly increased(P<0.05).After treatment,the levels of Wnt3 a and?-catenin were up-regulated(P<0.05).4.The key protein content of the jieguxujinwan treatment group was the highest,and the difference was statistically significant compared with other groups(P<0.05).Conclusion:1.jieguxujinwan can promote the fracture healing by promoting the activation and expression of Wnt/?-catenin signaling pathway and controlling the level of inflammatory factors.2.The efficacy of xunjinjieguwan compared with the control drug showed significant differences in the expression of inflammatory factors and Wnt/?-catenin key pathway proteins,showing obvious superiority.
